摘  要:目的 :研究显微分光光度计测量细胞DNA对肿瘤诊断和预后的价值。方法 :应用OPTON 0 3型扫描分光光度计和改良的DNA Feulgen染色方法对 15例涎腺发生的肿瘤、17例恶性淋巴瘤、2 3例早期胃癌和 2 0例食管及贲门早期浅表癌的癌细胞DNA含量进行了测量。测量结果与病理组织学诊断及随访结果进行了对比分析。结果 :癌细胞DNA含量增高 ,胞核DNA含量分布广泛 ,出现非整倍体细胞核。非整倍体型早期食管癌的复发率较二倍体型早期食管癌高 (P <0 0 5 )。结论 :细胞光度术测量细胞DNA含量对鉴别良、恶性肿瘤可以提供客观依据。DNA倍体数对食管癌预后判断有一定价值。本研究建立的DNA Feulgen染色方法效果极佳。Objective: To study the value of cytophotometric determination of cell nuclear DNA in diagnosis and prognosis of tumor.Methods: The neuclear DNA content of tumor cells in 15 patients with salivary tumor,17 patients with malignant lymphoma,23 patients with early gastric cancer and 20 patients with early superficial esophageal cancer were measured by using an improved DNA Feulgen staining and under OPTON type Ⅲ microspectrophotometer.The results of measurement of DNA content were compared with the histopathological diagnosis and the follow up data.Results: The increased neuclear DNA content,widespread DNA distribution and aneuploid DNA pattern were found in malignant tumor cells.The recurrence rate of tumor was significantly higher in esophageal cancer with aneuploid DNA pattern than in diploid tumor ( P <0 05).Conclusion: Cytophotometric determination of DNA content could provide an objective basis for differential diagnosis of benign or malignant tumor.DNA ploidy may be a valuable parameter for prognostic evaluation of esophageal cancer.Moreover,the effect of DNA Feulgen staining improved by us is very good.
