胃癌组织中p53蛋白及Rasp21蛋白的表达及意义

摘　　要：目的:联合检测p53蛋白及Ras p21蛋白在正常胃组织及胃癌组织中的表达,探讨二者与胃癌分化程度及5年生存率间的关系。方法:应用流式细胞术(FCM)对13例正常胃组织及27例胃癌组织细胞进行p53蛋白及Ras p21蛋白含量的定量检测。结果:p53蛋白在正常胃组织的表达均为阴性,85.2%(23/27)的胃癌组织p53表达阳性,p53蛋白表达量(FI值)和p53蛋白阳性表达率与胃癌组织分化程度及5年生存率有明显相关性(P<0.01)。Ras p21蛋白在正常胃组织中表达均为阴性,71.45%的胃癌组织Ras p21蛋白表达阳性;胃癌组织Ras p21蛋白表达量(FI)和Ras p21蛋白阳性表达率与胃癌组织分化程度及5年生存率有明显相关性(P<0.05)。p53蛋白表达与Ras p21蛋白表达呈正相关(r=0.755)。结论:p53蛋白及Ras p21蛋白过表达与胃癌组织的分化程度及预后有明显相关性,p53蛋白及Ras p21蛋白的高表达与胃癌的发生、发展有密切关系,联合检测p53及Ras p21蛋白可作为判断胃癌恶性程度及预后的有效指标。Objective：To detect the expression of p53 and Ras p21 proteins in normal gastric tissue and gastric cancer,and to study their correlation with differentiation of gastric cancer and rate of 5 year survival.Methods： By using flow cytometry to detect the expression of p53 and Ras p21 proteins in normal gastric tissue and gastric cancer tissue.Results：The expression of p53 was negative in normal gastric tissue,23 of 27（85.2%） gastric cancer tissure were positive,p53 positive expression rate and content were correlated with differentiation of gastric cancer and rate of 5 year survival,（P〈0.01）.The expression of Ras p21 was negative in normal gastric tissue,over-expression was found in 71.45% gastric cancer tissue.Ras p21 positive expression rate and content were correlated with differentiation of gastric cancer and rate of 5 year survival,（P〈0.05）.There was positive corrlation between p53 proteins and Ras p21proteins（r=0.755）.Conclusion：The overexpression of p53 and Ras p21 proteins are correlated with differentiation of gastric cancer and progenosis.The over expression of p53 and Ras p21 proteins is closely associated with occurrence and development of gastric cancer.To detect p53 and Ras p21 proteins may be useful for evaluating gastric cancer differentiantion and progenosis.
