摘 要:背景与目的:研究表明,Hedgehog(HH)信号通路为调控胚胎发育过程的重要通路之一,其活化参与多种实体肿瘤的发生,而功能性跨膜蛋白受体Ptched1(PTCH1)等位基因的沉默是HH通路激活和肿瘤发生的关键。本研究旨在探讨PTCH1在人膀胱尿路上皮癌中的表达及其与膀胱癌发生的关系。方法:收集人膀胱尿路上皮癌56例及正常膀胱组织19例,采用Real-time PCR、免疫组化、蛋白质印迹法(Western blot)方法检测标本中PTCH1 mRNA和蛋白的表达情况以及分布规律,分析PTCH1蛋白的表达与膀胱尿路上皮癌的临床生物学特征的关系。结果:PTCH1 mRNA在膀胱尿路上皮癌中的表达明显低于正常膀胱组织(P<0.05)。PTCH1蛋白在膀胱癌中的表达比正常组织低(P<0.01)。PTCH1表达与患者性别和年龄无关(P>0.05)。低分级癌中PTCH1的表达率明显低于高分级癌(P<0.05),侵润性膀胱癌PTCH1的表达率明显低于表浅性膀胱癌(P<0.05)。结论:PTCH1在膀胱尿路上皮癌的表达下调,并可能与膀胱尿路上皮癌的发生、发展密切相关。Background and purpose: Studies have showed that Hedgehog (HH) signal pathway was one of the important molecular mechanisms for regulating embryo development, and its activation was involved in a variety of solid tumors. PTCH1 alleles silencing played a key role in HH pathway activation and tumorigensis. The aim of this experiment was to observe the expression of PTCH1 in urothelial carcinoma of the urinary bladder and to analyze the relationship between its expression and clinical characteristics. Methods: Fifty-six cases of human urothelial carcinoma of the urinary bladder and 19 cases of normal samples were used in this experiment and the expression of PTCH1 was detected by real-time PCR, immunohistochemistry and Western blot, respectively. Results: The expression of PTCH1 mRNA was significantly lower in urothelial carcinoma of bladder than that of normal examples (P〈0.05). The expression of PTCH1 protein in urothelial carcinoma of bladder was also lower than normal examples (P〈0.01). No correlation was found between the expression of PTCH1 protein and the clinic-pathological features of gender and age (/9〉0.05). PTCH1 protein expression in low-grade bladder was lower than high-grade tumors (P〈0.05). The expression of PTCH1 protein in superficial tumors was also significantly lower than invasive tumors (P〈0.05). Conelnsion: PTCH1 may act as tumor-suppressor in the urothelial carcinoma of urinary bladder. The expression of PTCH1 may be related with pathological grading and clinical stages of urothelial carcinoma of bladder.
