摘 要:背景与目的:食管癌是我国最常见的恶性肿瘤之一。肾母细胞瘤过表达基因(nephroblastoma overexpressed gene,NOV)NOV/CCN3在肿瘤的发生、发展中起重要作用。本文研究过表达NOV/CCN3对食管癌细胞增殖、迁移以及浸润的影响。方法:构建NOV/CCN3过表达质粒pEGFP-C1-NOV,利用脂质体转染的方法转染人食管癌细胞株ECa-109、SEG-1、BIC-1。用蛋白质印迹法(Western blot)技术检测NOV蛋白在实验组(转染pEGFP-C1-NOV质粒的ECa-109、SEG-1、BIC-1)和空载体组(转染pEGFP-C1质粒的ECa-109、SEG-1、BIC-1)的表达水平。通过计数细胞绘制细胞生长曲线,比较2组的细胞增殖差异。通过细胞划痕实验,比较实验组和对照组细胞迁移能力的差异。通过Transwell细胞侵袭实验,比较2组细胞的侵袭差异。结果:Western blot结果显示,实验组NOV蛋白的表达明显高于空载体组(P<0.05)。细胞生长曲线显示,实验组细胞生长速度比空载体组快(P<0.05);细胞划痕迁移实验显示,实验组细胞迁移速度显著高于空载体组(P<0.05)。细胞侵袭实验显示,实验组细胞穿越Matrigel基质胶的细胞数显著快于空载体组(P<0.05)。结论:NOV过表达可促进食管癌细胞的增殖,并促进食管癌细胞的迁移和侵袭。Background and purpose: Esophageal carcinoma is one of most common malignant tumors in China. Nephroblastoma overexpressed gene (NOV/CCN3) plays an important role in the occurrence and development of malignant tumors. The purpose of this research was to investigate the effects of overexpression of NOV/CCN3 on proliferation, migration and invasion of esophageal carcinoma cells. Methods: NOV/CCN3 overexpression plasmids were constructed and transfected into esophageal carcinoma cell line ECa-109, SEG-1 and BIC-1. Western blot was used to test NOV expression in experimental group cells (pEGFP-C1-NOV plasmid transfected ECa-109, SEG-1 and BIC- 1), empty vector transfected cells (pEGFP-C1 plasmid transfected ECa-109, SEG-I and BIC-1). The effects of NOV overexpression on proliferation, migration and invasion of experimental and empty vector group cells were analyzed by drawing growth curve, wound healing migration assay and Transwell matrigel invasion assay. Results: The NOV overexpressed esophageal carcinoma cells (ECa-109-NOV, SEG-1-NOV, BIC-1-NOV) showed increased growth rate compared to empty vector group (P〈0.05); Migration assay displayed that the NOV overexpressed esophageal carcinoma cells migrated faster than empty vector group; Transwell invasion assay displayed that the numbers of cells penetrated through matrigel membrane were significantly higher in NOV overexpressed cells compared to empty vector group cells (P〈0.05). Conclusion: Our data indicated that NOV plays an important role on increasing proliferation of esophageal carcinoma cells and it promotes migration and invasion of esophageal carcinoma cells.
