摘 要:转移是肿瘤致死的一个重要因素,其发生需要多基因参与、多步骤调控,相关基因及其作用机制的发现对肿瘤转移的预防与治疗具有重要意义。高迁移率族蛋白B1(high mobility group box 1,HMGB1)是一种非组蛋白染色质结合蛋白,在DNA复制、修复、重组和基因转录中起重要作用,在细胞损伤和炎症发生时释放至胞外参与信号转导,调节细胞的生长和分化等。近年研究表明,HMGB1在多种肿瘤细胞和组织中高表达,增强肿瘤细胞的增殖、侵袭和转移,促进肿瘤的发生、发展,提示HMGB1有可能成为治疗和预防肿瘤转移的靶标。现就HMGB1在肿瘤转移中的作用及其机制的研究进展作一综述。Metastasis is the major cause of death from cancer. Cancer metastasis is a complex multi-step process, in which multiple genes participate. This study aimed to find the related genes and understand their role and mechanisms would be crucial for prevention and treatment of cancer metastasis. High mobility group box 1 (HMGB 1) is a non-historic chromosomal protein, which is implicated in diverse biological processes including DNA replication, repair, recombination and transcription. During cell damage and inflammation, HMGB 1 can be released into the ml t to participate in extracellular signalling transduction, then regulate cell growth and differentiation. Recent studies showed that HMGB1 was highly expressed in many kinds of cancer cells and tissues, which promoted cancer development by enhancing cell proliferation, migration and invasion. These suggested that HMGB1 may represent a potential target for the therapeutic intervention in cancer. In this paper, we reviewed the role of HMGB1 in cancer metastasis and the mechanisms involved.
