摘  要:目的观察丹参对HSC-T6细胞基质分解素-1(MMP3)及其抑制因子基因表达的影响。方法以不同浓度的丹参(0.8、0.4g/L)作用于培养的肝星状细胞株HSC-T6细胞48h,收集细胞,提取总RNA;用逆转录-聚合酶链反应(RT-PCR)方法测定MMP3及基质金属蛋白酶组织抑制因子-1(TIMP-1)的基因表达水平。结果MMP3基因表达水平在丹参0.8、0.4g/L两组分别为2.03±0.32、2.19±0.26,而空白对照组为2.13±0.30。TIMP-1基因表达水平在丹参0.8g/L、0.4g/L两组分别为2.11±0.32、3.12±0.46,而空白对照组为4.03±0.56。结论丹参可明显抑制HSC-T6细胞TIMP-1的基因表达,且与剂量有关;但对MMP3的基因表达无明显影响。Objective To investigate the effects of salvia mihiorrhiza on gene expression of stromelysin-1 ( MMP3 ) and tissue inhibitor of metalloproteinase-1 ( TIMP-1 ) in HSC-T6 cells. Methods HSC-T6 cells were cultured in the medium containing different concentrations of salvia miltiorrhiza (0. 8 and 0.4 g/L) and collected after for 48 h. Total RNA of HSC-T6 cells was isolated and gene expression levels of MMP3 and T!MP-2 were measured by using reverse transcription polymerase chain reaction. Re- sults MMP3 gene expression levels in 0. 8 and 0.4 g/L salvia miltiorrhiza groups were 2. 03 ± 0. 32 and 2. 19±0.26 respectively, and those in control group was 2.13 ±0. 30. TIMP-1 gene expression levels in 0. 8 and 0.4 g/L salvia miltiorrhiza groups were 2. 11 ±0. 32 and3.12 ±0. 46 respectively, and those in control group was 4. 03 ±0. 56. Conclusion Salvia mihiorrhiza may significantly inhibit TIMP-1 gene expression in HSC-T6 cells in a concentration-dependent manner. Salvia miltiorrhiza has no effeet on MMP3 gene expression.
