复方藤梨根制剂对人胃癌裸鼠腹腔移植瘤NF-κB p65表达的影响

摘　　要：目的研究复方藤梨根制剂(FFTLG)对人胃癌裸鼠腹腔移植瘤的抑制效应及其与瘤组织核转录因子(NF-κBp65)表达的关系。方法建立人胃癌裸鼠腹腔移植模型。24只裸鼠随机分为4组:FFTLG高、中、低剂量组和生理盐水组,分别观察荷瘤裸鼠腹腔移植瘤的生长情况,检测NF-κB p65的表达。结果生理盐水组、FFTLG低、中、高剂量组的抑瘤率分别为0、7.8%、30.0%和33.3%,FFTLG各组与生理盐水组比较,差异均有统计学意义(P<0.05或P<0.01);腹腔瘤组织中NF-κB p65阳性表达细胞百分比分别为(39.67±19.82)%、(33.00±19.94)%、(29.20±17.68)%、(27.17±15.75)%。FFTLG组的中、高剂量组与生理盐水组比较,差异均有统计学意义(P<0.05)。结论 FFTLG对裸鼠腹腔移植瘤生长具有抑制作用,且随着剂量的增加其作用增强,其作用途径之一可能是通过下调NF-κB p65的表达。Objective To investigate the effects of Fufang TengliGen Mixture inhibiting human gastric cancer implanted in abdominal cavities of nude mice and the relation between anti-cancer effects of Fufang TengliGen Mixture and the expression of NF-κB p65.Methods Models of nude mice with human gastric cancer transplanted in their abdominal cavities were established.24 nude mice were randomly divided into 4 groups：groups by high,moderate,low thickness FFTLG mixture and group by NS,the growth of tumours in their abdominal cavities were observed,the expressions of NF-κB p65 in gastric cancer tissue were detected by immunohistochemistry,then the relation was discussed between them.Results The tumour inhibition rates of NS,low,moderate,high thickness FFTLG mixture groups were 0,7.8%,30.0% and 33.3%,respectively.Compared to control group,experiment groups all showed significant differences（P0.05 or P0.01）;expression rates of NF-κB p65 were（39.67 ±19.82）%,（33.00 ±19.94）%,（29.20 ±17.68）%,（27.17 ±15.75）%,respectively.Compared to control group,moderate,high thickness FFTLG mixture groups all showed significant differences（P0.05）.Conclusion FFTLG Mixture can inhibit the growth of tumours transplanted in abdominal cavities of nude mice,the more dosage,the better effect,its mechanism may partly be to lower the expression of NF-κB p65.
