摘 要:目的观察姜黄素纳米微粒对人胃癌MGC803细胞凋亡机制的影响。方法 MTT法建立细胞剂量效应曲线,光镜和电镜下观察细胞形态变化,流式细胞仪分析MGC803细胞周期改变,免疫组化法检测凋亡相关Bcl-2蛋白表达。结果 MTT法显示,姜黄素纳米微粒抑制胃癌MGC803细胞增殖,呈现剂量-时间依赖性;光镜观察姜黄素纳米微粒组细胞损伤较轻,电镜下核周隙清晰,突触小泡较多;流式细胞仪法显示,姜黄素纳米微粒使胃癌MGC803细胞阻滞于G2/M期;免疫组织化学法显示,姜黄素纳米微粒使凋亡相关Bcl-2蛋白表达下调。结论姜黄素纳米微粒诱导人胃癌细胞MGC803凋亡并且延长药物对胃癌MGC803细胞作用时间。Objective To observe the effects of curcumin nanoparticle on apoptosis of human gastric carcinoma cell line MGC803 in vitro.Methods MTT assay was used to establish the dose-effect curves.The changes of cells were observed by light microscope and electron microscope.Flow cytometry was used to analyze MGC803 cell cycle changes.Immunohistochemical staining was used for detecting expression status of apoptosis-related Bcl-2 protein.Results MTT results showed that curcumin nanoparticle could inhibit the proliferation of gastric carcinoma cell lines MGC803 in vitro,and it showed dose-time dependence.Damage of cell was lighter in the curcumin nanoparticle group,which were observed by light microscope,while electron microscope observation showed that perinuclear space of MGC803 cell was clear and had more synaptic vesicle.Flow cytometry showed that curcumin nanoparticle inhibited MGC803 cell in G 2 /M phase.Immunohistochemical staining showed that curcumin nanoparticle made the expression of Bcl-2 protein decrease in MGC803 cell. Conclusion Curcumin nanoparticle induces the apoptosis of human gastric carcinoma cell MGC803,which can prolong the drug action time against gastric carcinoma MGC803 cell.
