摘  要:目的通过分析β内酰胺耐药铜绿假单胞菌耐药基因与蛋白质表达情况,以此来研究铜绿假单胞菌发生耐药的机制。方法利用PCR技术扩增β内酰胺耐药铜绿假单胞菌耐药基因,通过双向电泳和质谱技术,分析β内酰胺耐药铜绿假单胞菌蛋白质表达的差异,并对差异显著的蛋白斑点进行分析。结果β内酰胺耐药铜绿假单胞菌可检测到VIM基因和TEM基因,但膜孔蛋白基因OPRD2则缺失,同时经过蛋白组学和指纹图谱分析,可见膜孔蛋白oprD2与vim和tem蛋白的点差异显著,与基因检测结果相一致。结论β内酰胺耐药铜绿假单胞菌中出现耐药基因VIM和TEM,同时高表达vim及tem蛋白;但OPRD2基因缺失,同时膜孔蛋白D2低表达,为进一步研究铜绿假单胞菌的耐药机制奠定基础。OBJECTIVE To research resistant genes and protein expression in pseudomonas aeruginosa with beta-lactam,so as to illustrate its resistant mechanism.METHODS The beta-lactam antibiotics resistant genes were identified by PCR,and protein expression was compared with standard pseudomonas aeruginosa.The differential proteins were measured and analysed by MALDI-TOF.RESULTS VIM,TEM and OPRD2 genes of pseudomonas aeruginosa with beta-lactam were identified.The changes of three types of proteins associated with beta-lactam drug resistance of pseudomonas aeruginosa were appeared.CONCLUSION The changes of genes and protein expression in pseudomonas aeruginosa with beta-lactam provide an impetus for further researches of the resistance of pseudomonas aeruginosa and the development of new treatment strategy.
