巴曲酶对局灶性脑缺血再灌注模型外周血单个核CD34^+细胞的影响

摘  要:目的探讨巴曲酶是否影响局灶性脑缺血再灌注模型的外周血单个核CD34+细胞的数量。方法采用改良大鼠大脑中动脉闭塞(MCAO)线栓法模型,将大鼠随机分为4组:对照组,假手术组,动物模型组,巴曲酶组。缺血2h后进行再灌注,取再灌注1、3、6h和2、3、4、7、14d,8个观察点,测外周血单个核CD34+细胞表达情况,比较巴曲酶组和模型组的异同。结果外周血单个核CD34+细胞:模型组与巴曲酶组1h~2d外周血单个核CD34+细胞数量较对照组均有减少,且模型组CD34+细胞较巴曲酶组高。再灌注后2d,模型组与巴曲酶组CD34+、CD34+细胞差异无统计学意义(P﹥0.05),而在3~7d明显呈下降趋势,且巴曲酶组CD34+细胞高于模型组。再灌注后7~14d,巴曲酶组与再灌注组外周血单个核CD34+细胞百分率又逐渐恢复到基线水平。结论巴曲酶对局灶性脑缺血再灌注模型的内皮祖细胞可能具有动员作用。OBJECTIVE To investigate the influence of batroxobin on the changes of circulating CD34 ^+ cells in cerebral ischemia /reperfusion rat model.METHODS A model of suture occlusion of right middle cerebral artery of male SD rats was used.90 mature male SD rats were randomly divided into 4 groups:control group,sham-operated control group,model group and batroxobin group.After right middle cerebral artery being occluded,the rats were suffered from ischemia for 2 hours before the reperfusion.1-hour(1h),3h,6h,2-day(2d),3d,4d,7d,14d were designed as the time points.There were five rats in each end point.Circulating CD34 ^+ cells were detected by cytoflowmetric analysis.Batroxobin group was compared with model group and control group.RESULTS The changes of Circulating CD34 ^+ cells were:In model group,there were mild decreasing of circulating CD34 ^+ cells from 1h to 2d after reperfusion in both of model group and batroxobin group with latter one lower than the former.There was no difference between batroxobin group and model group at 2d.Circulating CD34 ^+ cells sharply decreased from 3d to 7d after reperfusion with number of circulating CD34 ^+ in batroxobin group higher than model group,and returned to basal values at 14th day after cerebral reperfusion.CONCLUSION It suggests that the difference of the expression of CD34 is significant between batroxobin group and model group at various time points after reperfusion.Batroxobin might mobilize the endothelial progenitor cells in this model.
