摘  要:目的探讨神经营养素3(NT-3)对坐骨神经损伤大鼠肌萎缩的修复作用及其机制。方法 50只SD大鼠制成坐骨神经损伤模型,分为对照组和NT-3组,每组25只。显微镜下NT-3组右侧腓肠肌内注射NT-3质粒10μL(1 g.L-1),对照组注射生理盐水10μL。于注射后不同时间点观察2组大鼠caspase-3基因的表达情况、运动终板和肌细胞凋亡数目。结果坐骨神经损伤后大鼠caspase-3基因转录水平升高;NT-3组caspase-3基因转录水平和肌细胞凋亡率均低于对照组,差异有统计学意义(P<0.05);坐骨神经损伤后14、21 d NT-3组大鼠运动终板多于对照组,差异有统计学意义(P<0.05)。结论 NT-3抑制caspase-3基因的表达、肌细胞凋亡并提高运动终板数量,可能是促进神经纤维损伤后再生和减缓失神经肌萎缩的重要因素。Objective To study the restoration effect of neurotrophin 3 (NT-3) on amyotrophy after injury of sciatic nerve in rats and its mechanism. Methods Fifty adult healthy Sprague-Dawley (SD) rats were made model of sciatic nerve in- jury and randomly divided into NT-3 group and control group, twenty-five in each group. The rats in NT-3 group were given NT-3 plasinid 10 μL( 1 g·L-1 ) by intramuscular injection in right musculus gastrocnemius,while the control group was given the same amount of physiological saline. After injection,the expression of caspase-3 gene, the number of motor end plate and muscle cell apoptosis were detected at different time points. Results The transcriptional level of caspase-3 gene increased af- ter sciatic nerve injury. Compared with the control group, the transcriptional level of caspase-3 gene and the rate of muscle cell apoptosis in NT-3 group were decreased obviously, the differences were statistically significant( P 〈 0.05 ). Fourteen and twen- ty-one days after sciatic nerve injury ,the number of motor end plate in NT-3 group was higher than that in control group ,the difference was statistically significant ( P 〈 0.05 ). Conclusion NT-3 inhibits expression of caspase-3 gene and muscle cell apoptosis and improves the number of motor end plate that may be an important factor which could promote regeneration of nerve fiber and relieve denervated amyotrophy after sciatic nerve injury.
