摘  要:目的:探讨水杨酸钠对脂肪乳诱导的高游离脂肪酸大鼠胰腺组织氧化应激水平的影响。方法:分别给大鼠静脉输注脂肪乳,脂肪乳+水杨酸钠和生理盐水48h,检测胰腺组织中MDA、SOD含量及PDX-1mRNA的表达。结果:脂肪乳组胰腺组织MDA/SOD是生理盐水组1.6倍,水杨酸钠+脂肪乳组MDA/SOD较脂肪乳组下降30%。脂肪乳组PDX-1mRNA表达较盐水组减少30%,输注水杨酸钠使PDX-1mRNA表达升高20%。结论:游离脂肪酸增多使胰腺组织氧化应激反应增强,水杨酸钠可能通过减轻胰腺氧化应激,发挥保护胰岛β细胞作用。Objective:To investigate the effects of sodium salicylate on FFAs-induced rats and on oxidative stress in pancreas tissues.Methods: Intralipid(IH),Sodium Salicylate + Intralipid(SI),and saline(SAL) were intravenously infused for 48h in rats.The malondialdehyde(MDA) content,Superoxide Dismutase(SOD) activity and the expression of duodenal homeobox-1(PDX-1) mRNA in pancreas were examined.Results: Compared with SAL group,there was elevated 1.6 times of MDA/SOD in pancreas tissues in rats with IH,the ratio decreased 30% in SI group.Infusion of IH markedly decreased the express of PDX-1mRNA,which were obviously prevented by salicylate co-infusion.Conclusion: This study provides preliminary assessments of efficacy of sodium salicylate as a new treatment for FFAs-induced β cell dysfunction.The effect of decreasing β cell apoptosis by salicylate in part may be secondary to reduce the oxidative stress in pancreas.
