摘 要:目的:研究枸杞多糖对链脲佐菌素(STZ)致糖尿病大鼠的影响。方法:雄性SD大鼠50只腹腔注射STZ 40 mg·kg-1建立糖尿病模型。经检测空腹血糖(FBG)指数确定建立模型后随机分成5组:模型组、罗格列酮阳性组及枸杞多糖低、中、高剂量组,每组10只。ig给予枸杞多糖0.4,0.8,1.6 g·kg-1·d-1,阳性组给予罗格列酮0.4 g·kg-1·d-1,正常组及模型组则给予等量生理盐水,连续ig 30 d。进行行为学观察,采用葡萄糖氧化酶法测定FBG;放射免疫法测定空腹胰岛素(FNS)、胰岛素敏感指数(ISI)及胰岛素抵抗(IR);酶联免疫法(ELISA)测定大鼠血清瘦素(Leptin)、肿瘤坏死因子-α(TNF-α)和白介素-6(IL-6);RT-PCR测定胰岛组织诱导型一氧化氮合酶(iNOS)的表达。结果:与模型对照组比较,给药组明显缓解糖尿病大鼠病情,枸杞多糖显著降低糖尿病大鼠的血糖和有效恢复胰岛素水平(P<0.01),并显著降低血清中炎症因子的水平(P<0.01);此外,下调胰岛组织中iNOS mRNA的表达(P<0.01)。结论:枸杞多糖对糖尿病大鼠具有明显疗效,其机制可能与恢复糖尿病大鼠胰岛功能以及增强机体抗氧化应激能力和抑制脂毒性有关。Objective: To investigate the effect of wolfberry polysaccharide on diabetic rats induced by streptozotocin. Method: Fifty male SD rats were injected peritoneally by streptozotocin (STZ) with a dose of 40 mg .kg-ltoestablish diabetic rat models. Successful rats model confirmed by detecting fasting blood-glucose (FBG) were randomly divided into 5 groups: normal control group, model group, rogridone group (0. 4 g .kg-l-d-1 ), low-, medium-and high-dosage groups of wolPoerry polysaccharide (0.4, 0. 8, 1.6 g -kg-1-d-1). The rats in the normal and model control groups were given same volume of normal saline. The drugs were given to mice daily for 30 consecutive days. Glucose oxidase (GOD) method and radioimmunoassay (RIA) were used to examine the Fasting blood glucose (FBG) ; fasting insulin (FNS) , the insulin sensitivity index (ISI) and insulin resistance (IR) , enzyme-linked immunosorbent assay (ELISA) was used to determine serum leptin, tumor necrosis factor-or (TNF-a) and interleukin-6 (IL-6), iNOS expression in islet tissue was measured with RT-PCR assay. Result: Wolfberry polysaccharide significantly reduce the levels of blood-sugar, and effectively alleviate insulin levels (P 〈 0. 01 ) , reduce serum levels of inflammatory factor significantly (P 〈 0.01 ) and in addition, down-regulate iNOS mRNA expression in islets (P 〈 0.01 ). Conclution: The results suggest that wolfberry polysaccharide has the obvious hypoglycemic and hypolipidemic effects which is related to its improving antioxidant capacity and restoring islet function and inhibiting lipotoxicity.
