摘　　要：目的　研究 β 转化生长因子 (TGF β)在异种骨移植局部对肿瘤坏死因子 (TNF) αmR NA表达的调节作用。方法　将TGF β复合于牛松质骨载体中并植入小鼠股部肌肉内 ,在术后 4、7、14和 2 1d ,应用原位杂交技术检测植骨局部TNF αmRNA的表达与细胞定位 ,图像分析观察TNF αmRNA表达量的变化规律。结果　术后 4、7、14和 2 1d ,植骨局部均可检测到TNF αmRNA的表达。TNF αmRNA主要定位于载体周边的单核细胞、中性粒细胞和成纤维细胞 ,复合TGF β松质骨载体组 4、7、14和 2 1dTNF αmRNA表达量均明显低于未复合TGF β组 (P <0 .0 1)。 结论　在异种骨移植局部多种细胞表达TNF αmRNA ,TGF β在异种骨移植局部可下调TNF αmRNA的表达水平。Objective\ To study the regulation of transforming growth factor beta (TGF β) on the expression of tumor necrosis factor alpha (TNF α) mRNA in the area of xenogeneic bone implant. Methods\ The bovine cancellous bone particles containing TGF β were implanted into the muscle of the legs of mice, and the muscles were removed 4,7,14 and 21 days later. The expression of TNF α mRNA in the site of implant was assayed by in situ hybridization, and was analyzed by computer to find some trends. Results\ The expression of TNF α mRNA was detectable in the area of implantation at 4,7,14 and 21 days after implantation. TNF α mRNA was expressed mainly by monocytes, macrophages, neutrophils and fibroblasts. The expression of TNF α mRNA in the bone particles containing TGF β was much lower than that of particles not containing TGF β ( P < 0.01 ). The peak of expression was at 14th day. Conclusion\ Many different kinds of cells express TNF α mRNA in the area of xenogenic bone implant. The expression of TNF α mRNA could be downregulated by TGF β in the region of xenogeneic bone implantation.
