摘 要:目的应用H9c2心肌细胞系,力图发现雌二醇对缺氧状态下心肌细胞的钠泵活性及其β1亚单位表达的影响。方法使用0,1nm,100nm浓度的雌二醇分别作用正常培养状态和缺氧培养的H9c2细胞5min,10min,20min、30min作为雌二醇对钠泵活性的短效作用研究;使用0,0.01nm,1nm,100nm和1μm的雌二醇作用24h,作为雌二醇对钠泵活性及其β1亚单位表达影响的长效作用研究。结果两种浓度的雌二醇不存在对钠泵活性的短效作用。缺氧能显著降低钠泵活性及其β1亚单位的表达(P<0.05),同时100nm和1μm的雌二醇能有效逆转上述作用(P<0.05)。结论钠泵活性和表达的降低可能参与了缺氧状态下心肌细胞的病理生理改变,雌二醇可能对缺氧心肌细胞具有保护作用。Objective Investigate the effect of estrogen on the activity and expression of Na pump in cardiac cells under hypoxic condition using H9c2 cells.Methods For the short-term effect research of estrogen,0,1nm and 100nm estrogen was used in the culture of H9c2 cells under hypoxic and normal condition.For the long-term effect research of estrogen,0,0.01nm,1nm,100nm and 1μm estrogen was used in the culture of H9c2 cells under hypoxic and normal condition.Both of the activity and expression of Na pump was determined in these two groups.Results The estrogen of 1nm and 100nm has no short-term effect the activity and expression of Na pump in H9c2 cells under hypoxic and normal condition.The activity and expression of Na pump were significantly lower under than hypoxic condition that of normal condition(P0.05),and this effect could be reversed by 100nm and 1μm estrogen(P0.05).Conclusion The decrease of the activity and expression of Na+,K+-ATPase may take part in the pathology change of cardiac cells,and estrogen may has the protect effect to the cardiac cells under hypoxic condition.
