摘  要:目的观察血管瘤来源的间充质干细胞的体内分化结果。方法应用贴壁筛选法从增生期血管瘤中分离间充质干细胞,将血管瘤来源的间充质干细胞/Matrigel混合物注射于裸鼠背部皮下,定期取出移植块,应用HE染色、免疫组化染色等方法观察移植块的病理结构。结果血管瘤来源的间充质干细胞/Matrigel移植块的中心区域在1周后可见血管形成,移植块的边缘区域可见少数脂肪细胞形成。随着时间的延长,中心区域的微血管逐渐减少消失,演变为纤维样组织;移植块边缘区域的脂肪细胞数量逐渐增多。结论血管瘤来源的间充质干细胞的体内分化过程与血管瘤的病理演变过程类似,提示血管瘤可能是来源于间充质干细胞的特殊肿瘤。Objective To observe the results of differentiation of hemangiomaderived mesenchymal stem cells (Hem-MSCs) in vivo. Methods MSCs were isolated from hemangioma in the proliferating phase by their selective adhesion to plastic culture dishes. Hem-MSCs/Matrigel mixture was injected subcutaneously into the back of nude mouse. These Hem-MSCs/Matrigel plugs were harvested at 1, 2, 4, and 8 weeks, then were fixed in 4% formaldehyde, and paraffin embedded. HE staining and immunohistochemistry staining were performed to observe the structure of the plugs. Results Vasculogenesis/angiogenesis were observed in the central area of the plugs at lweek, and adipogenesis in the marginal area. In the plugs at 2, 4, and 8 weeks, the microvessels in the central area decreased continuously, and regressed into fibrous tissue finally. The adipocyte in the marginal area increased continuously. Conclusion The in vivo differentiation of Hem-MSCs is similar to the pathological evolution of infantile hemangioma, which suggests that hemangioma may be a specific tumor from MSCs.
