摘  要:目的:实验观察灌胃天麻水提取物对大鼠下丘脑和肾上腺组织酪氨酸羟化酶(TH)和多巴胺-β-羟化酶(DBH)遗传因子的表达影响,探讨天麻的镇静作用机制。方法:分别取不同剂量组(10、20、40、80 mg/kg灌胃)和不同时间组(灌胃后2、4、6、8 h)大鼠的下丘脑和肾上腺组织,通过逆转录-聚合酶链反应(RT-PCR)与相应对照组(生理盐水灌胃)比较TH及DBH遗传因子的表达。结果:3个剂量组(20、40、80 mg/kg)大鼠下丘脑组织的TH及DBH遗传因子的表达与对照组相比均明显降低(为P<0.05,P<0.01,P<0.01)。肾上腺组织的TH表达在4个剂量组(10、20、40、80 mg/kg)与对照组相比都明显降低(P<0.05,P<0.05,P<0.01,P<0.01);DBH表达在3个剂量组(20、40、80 mg/kg)与对照组相比都明显降低(P<0.05,P<0.01,P<0.01)。天麻提取物灌胃(40 mg/kg)4小时及6小时后,下丘脑及肾上腺组织的TH和DBH遗传因子的表达与对照组相比均明显降低(均为P<0.01)。结论:天麻可降低下丘脑和肾上腺组织的酪氨酸羟化酶及多巴胺-β-羟化酶的活性。Objective:To investigate the effects of the orally administered aqueous extracts of Gastrodia Elata (GE) on the ex- pression levels of Tyrosine Hydroxylase (TH) and dopamine-[3- hydroxylase (DBH) mRNAs in hypothalamus and adrenal gland of rats, and its possible mechanism of sedation. Methods:Rats were randomly divided into a control group, and four different dose GE groups ( 10, 20, 40, 80mg/kg of GE), and 4 time - duration GE groups (2h, 4h, 6h, 8h after administration of 40 mg/kg of GE), and the expression levels of TH and DBH mRNAs in hypothalamus and adrenal gland were analyzed using RT - PCR. Results: Com- pared with the control, different dose GE significantly decreased ( P 〈 0.05 or P 〈 O. 01, except for 10 mg/kg) the expression levels of TH and DBH mRNAs in hypothalamus, and all dose groups of GE significantly decreased the expression of TH (P 〈 0.05 or P 〈 O. 01 ) , and the expression of DBH (P 〈0.05 or P 〈0.01, except for 10 mg/kg) in adrenal medulla. 4 and 6 h after administration of GE significantly decreased the expression levels of TH and DBH mRNAs in hypothalamus and adrenal medulla ( P 〈 0.01 ). Conclu- sions : GE could decrease the enzymatic activity of TH and DBH in hypothalamus and adrenal medulla of rats.
