摘 要:目的研究琥珀酸酯改性白及多糖的抗氧化活性。方法分别采用Fenton法和邻苯三酚自氧化法测定不同取代度(DS)白及多糖琥珀酸酯BT-2(DS=0.38)、BT-3(DS=0.69)、BT-4(DS=0.85)对OH.和O2-.的抗氧化活性。结果白及多糖琥珀酸酯对OH.的半数清除率浓度(mg/ml)依次为1.48±0.01(BT-4)、2.65±0.01(BT-3)、2.79±0.02(BT-2);对O2-.的半数清除率浓度(mg/ml)依次为1.38±0.06(BT-4)、6.44±0.02(BT-3)、21.33±0.13(BT-2)。结果显示,不同取代度白及多糖琥珀酸酯对OH.和O2-.的清除能力大小依次为BT-4>BT-3>BT-2(P<0.05)结论白及多糖琥珀酸酯的抗OH.和O2-.活性随取代度增大而显著增强。Objective To study the antioxidant activities of succinated glucomannan from Bletilla striata.Methods Antioxidant activities of succinated glucomannan with different degree of dubstitution(DS) including BT-2(DS=0.38),BT-3(DS=0.69) and BT-4(DS=0.85) against OH·and O-2·were measured by Fenton reaction and autoxibation of pyrogallic acid.Results The IC50(mg/ml) of succinated glucomannans against OH· was measured to be 1.48±0.01(BT-4),2.65±0.01(BT-3),2.79±0.02(BT-2);while their IC50(mg/ml) against O-2·was determined to be 1.38±0.06(BT-4),6.44±0.02(BT-3),21.33±0.13(BT-2),respectively.Conclusion The antioxidant activities of succinated glucomannans against OH·and O-2· were increased apparently with increasing DS.
