幽门螺杆菌对胃癌BGC-823细胞中Bcl-xl基因表达的影响及作用

摘　　要：目的探讨幽门螺杆菌超声裂解液对胃癌BGC-823细胞中Bcl-xl基因表达的影响,以及Bcl-xl基因在胃癌BGC-823细胞增殖中所起的作用。方法分别用东亚型和西方型幽门螺杆菌超声裂解液处理胃癌BGC-823细胞24h,MTT法检测细胞增殖情况,荧光定量PCR法检测胃癌BGC-823细胞中Bcl-xl基因mRNA水平的表达情况。设计特异的Bcl-xl shRNA阻断Bcl-xl基因的表达,抑制胃癌BGC-823细胞增殖。结果螺杆菌超声裂解液处理胃癌BGC-823细胞24h后,与对照组相比,处理组BGC-823细胞出现增殖(P<0.01),BGC-823细胞中Bcl-xl基因mRNA水平的表达也出现上调;并且东亚型螺杆菌超声裂解液处理组的促增殖作用强于西方型处理组(P<0.01),东亚型螺杆菌超声裂解液处理组细胞中Bcl-xl基因mRNA的表达水平也高于西方型处理组(P<0.01)。Bcl-xl shRNA可以下调Bcl-xl基因mRNA及蛋白水平的表达,同时抑制胃癌BGC-823细胞的增殖。结论 Bcl-xl基因对幽门杆菌感染相关性胃癌中癌细胞的增殖起了促进作用。Objective To investigate the effect of helicobacter pylori（Hp） extract on cellular proliferation and the expression of Bcl-xl mRNA in human gastric cancer BGC-823 cell lines, and to investigate the role of Bcl-xl expression on the proliferation of BGC-823 cells. Methods Human gastric cancer BGC-823 cell lines were treated with extracts from the East Asia type and Western type of Hp for 24 h. Cell proliferation were evaluated by MTT. The mRNA levels of Bcl-xl in BGC-823 cell were detected by real time quantitative PCR（RT-PCR）. Bcl-xl -mediated RNA interference technique was employed to inhibit the expression of Bcl-xl gene and the proliferation of BGC-823 cells,the levels of Bcbxl gene was inhibited by Bcl-xl shRNA. Results The proliferation of BGC-823 cell treated with Hp extract was observed 24 hours later,magnitude of enhancement of cellular proliferation in the East A sia type was higher than that in the Western type（P〈0.01）. The levels of Bcl-xl mRNA showed higher in human gastric cancer BGC-823 handled with Hp extract （P〈0.01）, compared to the control group. Statistical significance of the expression of Bcl-xl mRNA was also found between the East Asia type group and the Western type group（P〈0.01）. Bcl-xl shRNA could significantly reduced the expression of Bcl-xl mRNA and protein and the proliferation of BGC-823 cells. Conclusion It indicated that the prolif eration of Hp-related gastric cancer was promoted by Bcl-xl gene.
