摘  要:目的探讨caveolin-1和cathepsin D蛋白在非小细胞肺癌(non-small cell lung carcinoma,NSCLC)中的表达及其与微血管密度(microvessel density,MVD)及临床病理特征之间的关系。方法用免疫组织化学法检测caveolin-1和cathepsin D在86例NSCLC和20例癌旁肺组织中的表达,以及CD34在NSCLC中的表达,并对MVD计数。结果 caveolin-1在NSCLC组织中表达率显著低于癌旁肺组织(P=0.000);而cathepsin D在NSCLC组织中表达率显著高于癌旁肺组织(P=0.024)。caveo-lin-1和cathepsin D的表达均与TNM分期(P1=0.001;P2=0.008)和淋巴结转移有关(P1=0.032;P2=0.001)。caveolin-1和cathepsin D阳性病例的MVD值明显高于阴性病例(P=0.000)。caveolin-1与cathepsin D在NSCLC中呈正相关(r=0.307;P=0.004)。结论 caveolin-1在肺癌中具有双重作用;caveolin-1、cathepsin D高表达可能刺激间质内微血管密度增高,共同促进肺癌的侵袭和转移。Objective To study the expression of caveolin-1 and cathepsin D in non-small cell lung carcinoma(NSCLC) and their correlation with microvessel density(MVD) and clinicopathologic parameters.Methods Immunohistochemistry staining was used to detect caveolin-1,cathepsin D expression in 68 NSCLCs and 20 adjacent noncancerous lung tissues and CD34 expression in NSCLC.MVD was detected by CD34 immunostaining.Results The positive rate of caveolin-1 protein in NSCLC was significantly lower than those in adjacent noncancerous lung tissues(P=0.000).On the contrary,the positive rate of cathepsin D in NSCLC was significantly higher than those in adjacent noncancerous lung tissues(P=0.024).Caveolin-1 and cathepsin D expression were both correlated with higher TNM stage(P1=0.001;P2=0.008) and lymph node metastasis(P1=0.032;P2=0.001).MVD in the positive expression of caveolin-1 and cathepsin D were higher than those in the negative cases(P=0.000).The expression caveolin-1 was positively correlated with the expression of cathepsin D in the NSCLC(r=0.307;P=0.004).Conclusion caveolin-1 may have a dual effect in the NSCLC.High expression of caveolin-1 and cathepsin D may play a vital role in the progression of NSCLC and closely correlate with tumor angiogenesis.
