摘 要:鱼鳞中含有丰富的蛋白质资源,如何充分利用该资源越来越受到关注.利用碱性蛋白酶水解海洋鱼鳞蛋白制备鱼鳞蛋白多肽.通过考察温度、时间、酶量、底物浓度等单一因素对水解鱼鳞蛋白的影响,确定并设计了L9(34)正交试验因素和水平,最终获得了碱性蛋白酶水解海洋鱼鳞蛋白优化工艺条件,即温度60℃,时间1.5h,酶量3%(质量分数),底物质量分数10%.在此条件下,酶解所得水解度为32.38%.与其他方法比较,该工艺条件下水解时间大大缩短.检测了该条件所得蛋白肽段的分子质量分布,结果表明酶解产物主要集中在1~6ku.由此可见,该优化条件适合规模化生产,所得蛋白肽可以满足食品、化妆品行业的需求.Protein is very abundant in Sea fish scales. How to utilize it is now concerned more and more. Here sea fish sales were hy drolyzed to prepare peptides using alkaline protease. The irifluence factors including temperature,time, enzyme amount and substrate concentration were investigated and their levels were also determined for designing orthogonal experiment to optimize the condition. Based on the research work,the optimum treatment condition was summarized as follows:temperature 60 ℃ ,hydrolysis time 1.5 h, enzyme amount 3 % (by mass) and substrate concentration 10 %(by mass), under which degree of hydrolysis (DH) could be obtained at 32.38%and the reaction time was shortened greatly compared with previous report. Molecule weight distribution of peptides from this hydrolysis process was also investigated and the result was about 1-6 ku. The result indicated that the conditions weresuitable for large-scale production and molecule weight of peptides can meet the requirement from food and cosmetic industry.
