脂联素基因标签单核苷酸多态性与非酒精性脂肪肝病的相关性

摘　　要：目的探讨脂联素基因多态性与非酒精性脂肪性肝病(NAFLD)遗传易感性的关系。方法从2005年及2009年广东省汉族人群NAFLD流行病学调查的人群中筛选出2次调查均为NAFLD的119例患者为NAFLD组,2次调查均正常的350例为对照组,应用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)方法检测脂联素7个标签单核苷酸多态性:rs182052、rs16861205、rs822396、rs7627128、rs1501299、rs2241767、rs3774261,用多因素logistic回归分析危险因素,并用SHEsis软件进行单体型分析。结果 rs2241767的GG基因型是NAFLD患病的危险因素(OR=7.471,95%CI为2.836～19.684,P<0.001),rs1501299的TT基因型是NAFLD患病的危险因素(OR=5.040,95%CI为1.919～13.236,P=0.001),rs3774261的GG基因型是NAFLD患病的危险因素(OR=3.546,95%CI为1.402～8.972,P=0.008),单体型分析结果显示,ATA单体型是NAFLD的保护因素(OR=0.144,95%CI为0.037～0.558,P=0.001),GGA单体型是NAFLD的危险因素(OR=12.086,95%CI为2.422～60.302,P<0.01),GTG单体型是NAFLD的危险因素(OR=15.355,95%CI为3.946～59.748,P<0.001)。结论 rs2241767、rs1501299、rs3774261基因多态性与NAFLD遗传易感性相关。Objective To investigate the correlation between adiponectin gene polymorphisms and susceptibility of non- alcoholic fatty liver disease （NAFLD）. Methods From 2005 to 2009, 350 normal controls and 119 patients with persistent NAFLD were enrolled in Han population in Guangdong province epidemiological follow - up survey. The single nucleotide polymorphisms （SNP） of seven tagging single nueleotide polymorphism （tagSNP） of adiponectin, inclu- ding rs182052, rs16861205, rs822396, rs7627128, rs1501299, rs2241767, and rs3774261, was detected using polyme- rase chain reaction - restriction fragment length polymorphism （ PCR - RFLP）. Multivariate regression logistic analysis was applied to identify the risk factors of NAFLD, while SHEsis software was applied for haplotypes analysis. Results The GG genotype of rs2241767, Tr genotype of rs1501299, and TT genotype of rs1501299 were independent risk factors of NAFLD （ OR =7. 471, 5. 040, 3. 546, respectively, P 〈0. 001 ）. The concordant results were obtained in haplotypes analy- sis. Conclusion The polymorphisms of 2241767, rs1501299 and rs3774261 were associated with NAFLD susceptibility.
