摘 要:目的检测缺氧诱导因子-1α(Hypoxia-inducible transcription factor-1α,HIF-1α)、赖氨酰氧化酶(lysyloxidase,LOX)和PINCH(particularyinteresting new cysteine-histidine rich protein)蛋白在人喉鳞状细胞癌(简称鳞癌)中的表达,探讨三者在喉鳞癌发生发展、转移及预后的意义。方法采用免疫组织化学方法检测38例喉鳞癌组织中HIF-1α、LOX及PINCH的表达情况,并分析其表达的相关性。结果①HIF-1α、LOX和PINCH在喉鳞癌组织中表达显著高于癌旁黏膜组织;②HIF-1α和PINCH在淋巴结转移组与无转移组表达阳性率分别为100.0%、100.0%和67.7%、70.8%,统计学差异具有显著性意义;LOX在淋巴结转移组与无转移组表达,无显著性差异;③HIF-1α、LOX、PINCH的表达与患者的年龄、性别、原发部位、T分级及临床分期等临床病理因素无关;④HIF-1α在喉鳞癌中的表达与PINCH、LOX的表达存在相关性。结论 LOX、PINCH在人喉鳞癌中的表达存在相关性,可作为喉癌治疗的一个新靶点。HIF-1α的高表达可能是造成LOX、PINCH表达变化的重要影响因素之一。OBJECTIVE To detect the expressionof hypoxia inducible transcription factor-la (HIF-1α) , particulary interesting new cysteine-histidine rich protein (PINCH) and lysyloxidase (LOX) and their potential significances in laryngeal carcinoma. METHODS The expression of HIF-1α, PINCH and LOX in specimen of 38 patients with laryngeal carcinoma were detected by immunohistochemical method. There were 14 cases with and 24 cases without cervical lymph node metastasis. The normal laryngeal mucosal tissues were used as control. RESULTS 1.The expression level of HIF- 1α, PINCH and LOX was significantly higher in larynx squamous cell carcinoma tissues than in adjacent normal tissues; 2.Between the groups with and without cervical metastasis, significant statistically difference was existed in the expression of PINCH and HIF-1α There was stronger expression in group with cervical metastasis. however, the expression of LOX has no significant difference; 3.The expression level of HIF-I~, PINCH and LOX had no relationship with age, gender, clinical type, clinical stage or other clinical pathological factors; 4.The correlation was found between expression of HIF-1α and PINCH, also between expression of HIF-1α and LOX. CONCLUSION The expression of PINCH and LOX are regulated in laryngeal squamous cancer cells, and maybe correlated with the invasiveness and metastasis of the laryngeal cancer. The high expression of HIF-1α may play an important role in the regulation the expression of PINCH and LOX.
