摘  要:目的观察三氧化二砷(As2O3)对人大肠癌SW620细胞凋亡的影响,进一步研究As2O3作用后,凋亡相关基因FOX03a、Bcl-2和Bax表达的关系。方法体外常规培养人大肠癌SW620细胞,以不同浓度As2O3干预24h,观察细胞形态变化;Hoechst检测药物作用后细胞核形态变化;RT-PCR检测药物作用后FOX03a、Bcl-2和BaxmRNA的表达;SP法检测药物作用后FOX03a、Bcl-2和Bax蛋白的表达。结果As2O3作用后,SW620细胞数量明显减少,细胞变小、变圆,染色质固缩,胞核致密浓染,核碎裂增多并出现凋亡小体;FOX03a和BaxmRNA和蛋白的表达随As2O3浓度增加而增加,Bcl-2mRNA和蛋白的表达随As2O3浓度增加而减少。结论As2O3具有诱导大肠癌SW620细胞凋亡的作用,且与As2O3浓度有量效关系。其机制可能是通过上调FOX03a因子,继而上调Bax和下调Bcl-2的表达影响细胞凋亡。Objective To investigate cells apoptosis and the correlation of the expression of FOXO3a, Bcl -2 and Bax in colorectal cancer SW620 cells which is treated with arsenic trioxide. Methods Human colorectal cancer SW620 ceils were treated with different concentrations of arsenic trioxide, the morphological and structural changes of cells in each group were observed by microscope. The mRNA and protein expressions of FOXO3a, Bcl - 2 and Bax in human colorectal cancer SW620 cells were detected by RT- PCR and immunocytochemistry. Results In human colorectal cancer SW620 cells treated by arsenic trioxide, adherent ceils in each group gradually decreased, the difference of apop- totic morphology was obvious; The expressions of FOXO3a and Bax mRNA and protein were decreased. The expressions of Bcl -2 mRNA and protein were increased. Conclusion The results suggest that arsenic trioxide can induce colorectal cancer SW620 cells apoptosis, the mechanism may be related to increasing the expression of FOXO3a.
