摘  要:目的探讨牛磺酸对实验性糖尿病(DM)大鼠心脏的保护作用及其机制。方法将30只Wistar大鼠随机分为对照组(CN组)、DM组、牛磺酸组(TAU组)各10只,后两组均通过一次性尾静脉注射链脲佐菌素(STZ)制备DM模型;其后TAU组以牛磺酸300 mg/(kg.d)灌胃,CN组、DM组给予等体积生理盐水灌胃,连续8周。实验结束后,三组经腹主动脉取血检测丙二醛(MDA)、超氧化物歧化酶(SOD)、一氧化氮(NO)水平;取心肌组织,采用诱导型一氧化氮合酶(iNOS)检测试剂盒检测iNOS活性,采用TUNEL试剂盒检测细胞凋亡指数(AI)。结果与CN组比较,DM组血清MDA、NO水平增高(P<0.01、0.05),SOD活性下降(P<0.01),心肌组织iNOS活性增高(P<0.05),心肌细胞AI升高(P<0.01);与DM组比较,TAU组MDA、NO水平下降(P<0.05),SOD活性增高(P<0.05),心肌组织iNOS活性降低(P<0.05),心肌细胞AI明显降低(P<0.01)。结论牛磺酸对实验性DM大鼠心脏具有保护作用,可能机制为减轻氧化应激损伤、减少心肌细胞凋亡。Objective To investigate the cardioprotections and mechanism of taurine on experimental diabetes mellitus (DM) rats. Methods Thirty Wistar rats were randomly divided into control (CN)group, DM group and taurine (TAU) group with 10 in each, streptozocin(STZ) was administered to establish DM model in the latter two groups; Then taurine 300 mg/(kg d) was given to rats in TAU group, the same volume of saline was given to rats in CN and DM group, which lasted for 8 weeks. Then the productions of MDA and NO, the activities of SOD and iNOS were examined in all rats. Myo- cardium apoptosis index (AI)was assessed by terminal deoxyribonucleotidyl transferase mediated dUTP nick-end labeling assay. Results Compared with the CN group, productions of MDA and NO were .increased significantly ( P 〈 0.01 or O. 05 ) , the activity of SOD was decreased( P 〈 0.01 ) and the activity of myocardium iNOS was increased significantly, my- ocardium apoptosis was potentized in DM group (P 〈 0.01 ) ; Compared with the DM group, productions of MDA and NO were decreased significantly(P 〈 0.05 ), the activity of SOD was increased( P 〈 0.05 ) and the activity of myocardium iN- OS was decreased significantly (P 〈 0.05), myocardium apoptosis was also attenuated (P 〈 0.01 ) in TAU group. Conclu- sion Taurine can protect the experimental DM rats' heart by decreasing the injury of oxidative stress and lessening myocardium apoptosis.
