摘 要:利用14个家牛Y染色体特异性微卫星标记分析秦川牛、大别山黄牛和郧巴黄牛3个品种Y染色体多态性。UMN0920和UMN2303标记呈现不规则梯状条带,不适于研究中国黄牛Y染色体多态性。5个标记(UMN2908、UMN3008、UMN3007、UMN2001和UMN0504)呈现单态,另外7个标记(UMN0108、UMN2404、UMN0929、INRA189、UMN0905、UMN0103和UMN0803)表现多态,能用于中国黄牛Y染色体多态性分析。3个品种在7个多态标记的期望杂合度分别为0.412、0.442和0.470,多态信息含量分别为0.319、0.349和0.374,说明3个黄牛品种Y染色体上呈现中度多态。UMN2404和INRA189标记对所有公牛的瘤牛和普通牛单倍型鉴别率达到100%的一致,瘤牛和普通牛单倍型频率为17.3%和82.7%。瘤牛和普通牛单倍型频率在3个黄牛品种中频率不同,但普通牛单倍型为优势单倍型。Genetic diversity of Y chromosome in three Chinese indigenous cattle breeds (Qinchuan, Dabieshan and Yunba) were investigated by 14 microsatellite markers specific for cattle Y chromo- some. Two markers (UMN0920 and UMN2303) amplified irregular ladder-like bands, which were not suitable for investigating genetic diversity of Chinese cattle Y chromosome. Five markers (UMN2908, UMN3008, UMN3007, UMN2001 and UMN0504) were all monomorphic. Other seven markers (UMN0108, UMN2404, UMN0929, INRA189, UMN0905, UMN0103 and UMN0803) were all polymorphic, which can be used to study genetic diversity of Chinese cattle Y chromosome. The expected heterozygosity across these seven markers in the three breeds were 0. 412, 0. 442 and 0. 470 respectively; and the polymorphism information contents were 0. 319, 0. 349 and 0. 374 respec- tively, demonstrating a moderate degree of polymorphism in the Y chromosome of Qinchuan, Dabies- han and Yunba breeds. Consistently, UMN2404 and INRA189 markers displayed specific indicine and taurine haplotypes in each bull examined. The frequencies of indicine and taurine haplotypes were 17.3% and 82.7%, respectively. The taurine haplotype dominated in the three Chinese indigenous cattle breeds, although these frequencies varied amongst different breeds examined.
