摘  要:【目的】观察沙鼠脑缺血再灌注及美满霉素干预后,TGF-β3表达变化和高尔基体形态学变化,研究美满霉素的神经保护作用。【方法】50只沙鼠随机分为对照组、假手术组、缺血再灌注组、美满霉素干预组;建立颈总动脉阻塞脑缺血再灌注模型;HE染色观察存活神经细胞数;免疫组化法检测TGF-β3阳性细胞数及高尔基体形态学改变。【结果】缺血再灌注后TGF-β3表达水平升高,美满霉素减少缺血再灌注所致神经细胞死亡,并降低TGF-β3相应时间点的表达;仅缺血再灌注7d组中,部分高尔基体形态改变。【结论】美满霉素通过改变TGF-β3表达水平起到神经保护作用;美满霉素有助于维持高尔基体细胞器结构稳定。[Objective]To detect for neuroprotection mechanism of minocin in gerbils after cerebral ischemia. [Methods]Gerbils disease models of bilateral carotid artery occlusion (BCAO) have been established for 10 min. Fifty gerbils were randomly divided into four groups., normal control group(Group N), sham operation group (Group S), ischemia-reperfusion (I/R) group ( Group I/R) and minocycline intervention group (Group MI/R). In Group MI/R, minocin (45 mg/kg i. p, BID was given). The histological changes of cere- bral cells and the expressions of TGF-β3,Golgi apparatus(GA) were tested. [Results]After ischemia reperfu- sion, the expression of TGF-β3 increased. The morphological changes of some GA took place in Group I/R at 7- day-I/R. Minocin reduced the nerve Cell death caused by ischemia reperfusion, and dereased the expression of TGF-133 at corresponding time point. [ConclusionlMinocin may exert neuroprotection througth altering expression of TGF-β3 ;minocin is beneficial to the maintenance of the normal GA morphology.
