摘 要:目的甾体激素十一酸睾酮联合去氧孕烯和微量炔雌醇(TU/DSG/EE)对服用醋酸棉酚(GA)大鼠精子发生功能损伤恢复的影响。方法雄性Wistar大鼠随机分为TU/DSG/EE+GA、1%甲基纤维素+GA、Tu/DSG/EE和1%甲基纤维素(对照组)4组。TU/DSG/EE组大鼠持续服用TU/DSG/EE,对照组持续服用1%甲基纤维素,U/DSG/EE+GA组、1%甲基纤维素+GA组提前服用相应药物4周,再联合服用GA持续4周。各组停药恢复9周。服药4周、8周、停药恢复9周时进行大鼠血清和睾丸内激素水平;服药8周、停药恢复9周时对睾丸重量和睾丸精子头部计数检测及对大鼠睾丸生精上皮进行组织形态学分析。结果服药8周后,TU/DSG/EE+GA组的大鼠睾丸重量、睾丸精子头部计数分别为(0.3±0.04)g.0,1%甲基纤维素+GA组分别为(0.71±0.05)g、(24.2±3.3)×10^6个/克睾丸组织,TU/DSG/EE组分别为(0.53±0.06)g、(46.2±3.9)×10^6个/克睾丸组织,对照组分别为(1.66±0.08)g、(136.5±10.6)×10^6个/克睾丸组织,差异均有统计学意义。TU/DSG/EE+GA组的大鼠睾丸切片中未发现A级和B级生精小管,C级小管的比例(7.2%)显著地低于对照组(50.4%)、1%甲基纤维素+GA(19.6%)和TU/DSG/EE组(70.1%)。TU/DSG/EE+GA、TU/DSG/EE组的大鼠血清LH、FSH和睾丸间液睾酮(TIT)均显著低于对照组。恢复9周后,TU/DSG/EE+GA和TU/DSG/EE组的上述各项指标值均恢复到对照水平。结论激素可保护大鼠睾丸生精功能免受棉酚不可逆损伤进而促进其完全恢复,保护作用可能与睾丸内睾酮水平的降低密切相关。Objective To investigate the effect of additional treatment with testosterone undercanoate/desogestrel/ mini-dose ethinylestradiol (TU/DSG/EE) on recovery of spermatogenesis damaged by gossypol acetic acid (GA). Methods Adult male Wistar rats from groups of TU/DSG/EE + GA or GA were treated respectively with GA. combined with DSG/EE/TU or 1% methylcellulose (vehicle) for 4 weeks, then with the same dose of DSG/E/TU or vehicle for additional 4 weeks. These rats were allowed for a recovery peroid of 9 weeks. Control rats and rats of group H were given vehicle or DSG/EE/TU. Testicular weight, testicular sperm head counts, and histopathocological analysis of testis were applied to assess the spermatogenesis of treated rats after the 8-week treatment and the 9-week recovery. Results Testis weight, sperm head count, as well as mature spermatids and actively proliferating and differienting spermatogenic cells in testis of group TU/DSG/EE + GA rats were dramatically decreased after 8-week treatment when compared with those of rats from GA, H, or control groups. However, after 9-week recovery, the parameters mentioned above in rats from groups TU/DSG/ EE + GA and H returned to the levels similar to the controls. However, those of rats from group GA were still significantly different form the controls. Conclusion Additional hormonal treatment could protect the spermatogenesis of treated rats against destructive effects induced by gossypol acetic acid and thus stimulate its complete recovery.
