苦豆子凝集素粗品对9种常见致病菌的抑制效果

摘　　要：目的研究苦豆子种子凝集素粗品对几种常见细菌的抑制作用。方法获得不同硫酸铵饱和度下苦豆子种子凝集素粗品,用鸡血和鼠血红细胞分别检测其凝集活性,并以表皮葡萄球菌、粪肠球菌、臭鼻克雷伯菌等9种细菌作为供试菌,比较不同硫酸铵饱和度下凝集素粗品的抑菌活性。结果苦豆子种子凝集素粗品对鸡、鼠血红细胞有不同程度的凝集作用,对9种供试菌均有抑菌活性,其中对福氏志贺氏菌、肺炎克雷伯氏菌、类产碱假单胞菌和大肠埃希菌的抑制效果明显。结论结果表明苦豆子凝集素粗品具有抑制细菌生长的作用,其中对所选革兰氏阴性菌(G-)的抑菌效果好,本研究为苦豆子凝集素将来进一步开发成临床抗菌药物提供了实验依据。Objective To detect the inhibitory effects of lectins from Sophora alopecuroides seeds on several common bacteria.Methods The extraction of crude lectins from Sophora alopecuroides seeds was obtained after ammonium sulfate precipitation under different saturation and dialysis,and then its agglutinating activity was detected with erythrocytes of chickens and mice and antibacterial activity was measured by nine kinds of bacteria,such as,Staphylococcus epidermidis,Klebsiella penumoniae,Enterococcus faecalis,etc.Results The crude lectin showed different degrees of aggregation function on red blood cells of chickens and mice.The lectins had broadly inhibitory effects on the tested microbes,thereinto,the inhibitory effects on Shigella flexneri,Klebsiella pneumoniae,Pseudomonas pseudoalcaligenes and Escherichia coli were obvious.Conclusions These findings indicate that the crude lectins of Sophora alopecuroides display inhibitory effects on all the tested bacteria.Meanwhile,the inhibitory activities for gram negative bacteria are remarkable.This result should provide the experimental basis for the further development of clinical antibiotics of Sophora alopecuroides lectins in the future.
