摘 要:以沥窖污水处理厂的活性污泥作为菌种来源,通过实验室构建的序列间歇式反应器(SBR),从活性污泥多种微生物种群中,驯化、分离、筛选出一株能以苯胺黑药为惟一碳源的菌株AAF039,并对其进行了生理生化、16SrRNA基因序列分析;通过控制菌株生长和降解的底物含量、温度及pH,并以菌株生长和降解苯胺黑药的效果为依据,得出菌株AAF039的最适生长、降解条件;测定培养过程中营养液的CODCr与苯胺黑药剩余量,对苯胺黑药的生物降解过程进行初步推测.结果表明:菌株为死亡谷芽孢杆菌(Bacillus vallismortis),其生长能以苯胺黑药为惟一碳源;生长和降解苯胺黑药的最佳条件为底物质量浓度300mg/L、温度35℃,pH7.0,21h后,对苯胺黑药的降解率可达89.11%;苯胺黑药并不能彻底被生化降解,降解过程中产生了难以被菌株利用的中间产物.Activated sludge which came from Lijiao Sewage Plant was taken as bacterium source and established the SBR (sequencing batch reactor) system in laboratory. Bacterial strain AAF039 taken aniline aerofloat as only carbon source was acclimated, separated and sieved from multiple microbial population of activated sludge, and was analyzed by physiology and bi- ochemistry, and 16S rRNA gene sequencing. Through controlling growth, degrading substrate concentration, decreasing temperature and reducing pH value, the optimal condition for growth of bacterial strain AAF039 and biodegradation of aniline aerofloat was researched in terms of bacterial strain growths and aniline aerofloat degradation. Surplus of CODcr and aniline aero- float was measured in the process of cultivation, and the biodegradation process of aniline aero- float was conjectured. The results show: bacterial strain AAF039 is Bacillus vallismortis and uses aniline aerofloat as only carbon source; substrate concentration is 300 mg/L, temperature is 35 ~C and pH is 7.0, which are the optimal conditions of growth and degradation of aniline aerofloat and the degradation rate can reach 89.11% in 21 h;aniline aerofloat can not be degra- dated completely, and intermediate products generate in the process of degradation, which is hard to b~ used by baterial strain AAF039.
