摘 要:目的建立一种简单、便捷、有效检测K-ras突变的蝎子PCR新方法。方法同时用蝎子PCR法和Sanger测序法对87例结直肠癌组织的K-ras基因进行热突变型G12D(GGT>GAT)进行检测,统计其符合率。结果 87例结直肠癌组织样本中,蝎子PCR法和Sanger测序法检测到同样9例G12D突变,其符合率为100%。结论蝎子PCR法能有效检测到K-ras突变,且比Sanger测序法更为简便、快速,适合临床应用。Objective To establish a simple, convenient and effective scorpion PCR method to detect K-ras gene mutation. Methods K-ras gene mutation G12D(GGTGAT) of 87 colorectal cancer tissue samples were tested by scorpion PCR method and Sanger sequencing method. And coincidence rate was calculated. Results 9 cases G12D mutations were detected in 87 colorectal cancer tissue samples by both scorpion PCR method and sanger sequencing method. The coincidence rate was 100%. Conclusion The scorpion PCR method can detect K-ras gene mutation effectively. And it's more convenient and faster than Sanger sequencing method. It's very suitable for the clinical application in K-ras gene mutation detection.
