摘 要:目的观察脑络欣通对大脑中动脉闭塞缺血-再灌注(MCAO/R)大鼠血液流变学及相关调节因子的影响。方法将Wistar大鼠60只,随机分为假手术组、模型组、通心络组、脑络欣通低剂量(40 mg/ml)组、中剂量(80 mg/ml)组及高剂量(100 mg/ml)组,每组各10只。采用线栓法制备大脑中动脉闭塞缺血-再灌注(MCAO/R)模型。采用全自动血液流变仪检测全血黏度、血浆黏度、红细胞比容(HCT);高性能血凝仪检测血浆纤维蛋白原(FIB)含量;运用双抗体一步夹心法酶联免疫吸附实验,检测血小板生成素(TPO)、内皮素(ET)水平;运用激光多普勒仪,于造模后第14天监测各组大鼠的局部脑血流量(rCBF)。结果①假手术组、通心络组、脑络欣通3个剂量组大鼠的全血黏度各项、血浆黏度、HCT和FIB指标均低于模型组,差异有统计学意义,P<0.01。②通心络组、脑络欣通中剂量和高剂量组的HCT低于脑络欣通低剂量组,差异有统计学意义,P<0.01。③假手术组、通心络组及脑络欣通3个剂量组大鼠的TPO、ET水平均低于模型组,rCBF高于模型组,差异具有统计学意义,P<0.01或P<0.05。④脑络欣通中剂量组大鼠TPO水平低于脑络欣通低剂量组,rCBF高于脑络欣通低剂量组,差异具有统计学意义,P<0.05。结论脑络欣通可以改善MCAO/R模型大鼠的血液流变学指标,调节凝血纤溶系统相关因子,对缺血性脑血管病可能具有良好的作用。Objective To observe the effects of Naoluoxintong on hemorheology (rCBF) and related regulatory factors in rats with middle cerebral artery occlusion-reperfusion (MCAO/R) . Methods Sixty Wistar rats were randomly divided into sham operation, model, tongxinluo, naoluoxintong 40 mg,/ml, Naoluoxintong 80 mg/ml, and Naoluoxintong 100 mg/ml groups. A rat model of MCAO/R was induced by the intraluminal suture method. Whole blood viscosity,plasma viscosity and hematocrit (HCT) were detected by an automatic hemorheology analyzer. The content of fibrinogen (FIB) was detected by a high performance coagulation analyzer. The levels of thrombopoietin (TPO) and endothelin (ET) were detected by double antibody sandwich enzyme-linked immunosorbent assay (ELISA). The rCBF of the rats was detected by laser Doppler f/owmetry at the 14th day after modeling in each group. Results ①Whole blood viscosity, plasma viscosity, HCT, and FIB in the sham operation, Tongxinluo and Naoluoxintong groups were all lower than those in the model group. The differences were statistically significant ( P 〈 0.01 ). ②HCT in the Tongxinluo, Naoluoxintong 80 mg/ml and Naoluoxintong 100 mg/ml groups was lower than that in the Naoluoxintong 40 mg/ml group. The differences were statistically significant ( P 〈 0.01 ). ③The levels of TPO and ET in the sham operation,Tongxinluo and 3 Naoluoxintong groups were lower than those in the modelgroup, but the rCBF was higher than that in the model group. The differences were statistically significant ( P 〈 0.01 or P 〈 0.05 ). ④The level of TPO in the naoluoxintong 80 mg/ml group was lower than that in the naoluoxintong 40 mg/ml group, but the rCBF was higher than that in the naoluoxintong 40 mg/ml group. The difference was statistically significant ( P 〈 O. 05 ). Conclusion Naoluoxintong can improve the hemorheology indexes and regulate the relevant factors of the fibrinolysis coagulation system in MCAO model rats ,which has a good effect on ischemic cerebr
