摘  要:Shelterin复合体是端粒结合蛋白的核心成分,他的复杂调控与端粒酶、端粒延长替代途径(ALT机制)在保护端粒功能中发挥重要的作用.端粒是包含一段重复序列的DNA-蛋白复合体,通过端粒酶募集到染色体末端,稳定染色体末端,从而防止染色体融合.端粒的保护与基因组的稳定密不可分,而基因组的不稳定是肿瘤形成的分子基础.目前,恶性肿瘤已成为危害人类健康的主要疾病,其中消化系恶性肿瘤的发病率和死亡率均位居前列.许多研究表明端粒结合蛋白在胃癌、大肠癌和肝癌等消化系肿瘤中发挥重要作用.本文着重总结Shelterin复合体在消化系恶性肿瘤中的作用,为消化系恶性肿瘤的预防及靶向治疗提供重要依据.The Shelterin complex is the crucial components of telomere binding proteins.The regulation of this complex,together with telomerase and the alterative lengthening of telomeres(ALT mechanism),plays a critical role in maintaining telomere functions.Telomeres are DNA-protein complexes that contain short repeat sequences added on to the ends of chromosome by the telomerase for protecting the ends of chromosome and preventing chromosome fusion.The loss of protective function of telomeres is closely related to genome instability,and this is the molecular basis for tumor development.Thus,telomeres play key roles in the process of malignant tumor development.Many studies have shown that telomere binding proteins are associated with gastric,colorectal and liver cancers,and other digestive system tumors.This review will focus on the role of the shelterin complex in digestive system neoplasms to provide an insight into prevention and targeted therapy of these malignancies.
