摘 要:目的观察蜕皮甾酮(ecdysterone,EDS)对大鼠脑损伤的治疗作用。方法 SD雄性大鼠120只,按随机数字表法分6组,每组20只。采用Feeney法自由落体打击鼠脑造模。假手术组动物只开颅,不打击;治疗组:分别采用4、8、16 mg/kg的EDS腹腔注射模型动物;阴性治疗对照组:模型动物腹腔注射生理盐水;阳性治疗对照组:模型动物腹腔注射3 mg/kg依达拉奉。伤后7 d计算各组动物死亡率;检测各组动物脑组织含水量及丙二醛(MDA)、羟自由基能力及超(过)氧化物歧化酶(superoxide dismutase,SOD)等。HE染色观测各组大鼠脑组织海马病变情况。结果伤后7 d,16 mg/kg EDS治疗组的死亡率、脑组织含水量MDA与羟自由基含量较阴性治疗对照组明显降低(P<0.05),SOD活性显著升高(P<0.01),脑细胞损害明显减轻,大鼠海马CA1区正常神经元数量明显优于阴性治疗组(P<0.01)。结论蜕皮甾酮对脑损伤具有保护治疗作用。Objective To investigate the effect of ecdysterone (EDS) on traumatic brain injury (TBI) in rats. Methods A total of 120 male Sprague-Dawley rats were randomly and equally divided into 6 groups, sham-operation group, TBI group, edaravone treatment group and EDS treatment groups. Modified Feeney' s method was used to establish the rat model of experimental TBI with an impact of a weight of 30 g from 20 cm after craniotomy. Rats of EDS treatment groups were given EDS at 4, 8, and 16 mg/kg via perito- neal injection for 7 d. The other 3 groups were treated with normal saline or edaravone (3 mg/kg). The animal death rate was calculated, and the changes of the brain water content, malondialdehyde ( MDA), and superox- ide dismutase (SOD)in brain tissue were calculated and measured. Hematoxylin-eosin (HE) staining was used to observe the pathological changes of brain hippocampus tissue. Results EDS ( 16 mg/kg) resulted in significant decreases in the death rate, brain water content, and MDA content (P 〈 0.05 ), and an obvious increase in SOD content (P 〈0.01) compared with TBI group. EDS (16 mg/kg) also resulted in lesser damage in the brain cells, with the pyramidal cells number significantly higher than TBI group ( P 〈 0.01 ). Conclusion EDS exerts therapeutic effect on TBI through inhibiting free radical damage and brain edema.
