摘 要:目的评价不同种植体支抗材料镍铬合金、钴铬合金、钛和聚乳酸酯对人成骨样细胞MG-63的细胞毒性及护骨素(Osteoprotegerin,OPG)与OPG配体即破骨细胞分化因子(Receptor activator nuclear factor-kappa Bligand,RANKL)mRNA的表达量的影响。方法选择人成骨样细胞系MG-63作为细胞毒性的实验对象,分为5组,即对照组,浓度为0.05%的镍铬、钴铬、聚乳酸酯、钛微粒各一组。用MTT法检测细胞存活率;用半定量RT-PCR检测在相同浓度的各种材料的作用下,人成骨样细胞内OPG/RANKL mRNA的表达变化。结果浓度为0.05%的不同粒子对人成骨样细胞MG-63的细胞毒性及OPG/RANKL mRNA的表达量的影响:(1)镍铬微粒对细胞生长有一定的抑制作用,钛微粒则有较强的抑制作用;钴铬微粒和聚乳酸酯微粒对细胞的增殖无明显的副作用。(2)镍铬微粒对细胞OPG的表达产生抑制作用,与对照组比较,差别有统计学意义;钴铬微粒和聚乳酸酯微粒对细胞OPG的表达无明显影响,与对照组比较差别无统计学意义;钛粒子诱导OPG在细胞中的表达量大大增加,与对照组比较差别具有统计学意义。(3)镍铬微粒可有效诱导人成骨样细胞中RANKL的表达,钴铬微粒对RANKL的表达具有一定的诱导效果,两者与对照组比较,镍铬微粒诱导效果更强,呈现显著性差异,钴铬微粒则呈现一般性显著;而钛微粒则显著增加RANKL的表达量;聚乳酸酯对人成骨样细胞中RANKL的表达则无明显的诱导作用,与对照组比较,差别无统计学意义。结论聚乳酸酯材料的细胞毒性最小,对OPG/RANKL mR-NA的表达量影响最小;钛次之;而镍铬、钴铬合金对OPG/RANKL mRNA的表达量有一定的影响。Objective To evaluate the cytotoxicity of different kinds of implant materials(nickel-chromium;cobalt-chromium;titanium;PLA) on human osteoblas-like cells(MG-63) and the effects on the expression of OPG/RANKL mRNA.Methods Human osteoblast-like cells MG-63 was chosen to evalute the cytotoxicity of different kinds of implant materials(nickel-chromium;cobalt-chromium;titanium;PLA) with 0.05% concentration respectively.The cells were divided into 5 groups.The viability of osteoblast was detected by MTT method.The expression of OPG/RANKL mRNA were examined by RT-PCR.Results(1)Compared to the control,MTT test demonstrated that NiCr had some inhibitory action on the growth of osteoblast,and titanium could obviously inhibit the cell viability of osteoblast,but not PLA and CoCr;(2)compared with control group,NiCr could obviously decrease the cell viability of osteoblast and existed significant difference;Compared to the control,the expression of OPG has no effect with PLA and HAP.The expression of OPG has no effect;titanium could induce the increase of the expression of OPG in osteoblast,and existed significant difference;(3)NiCr and CoCr could induce the expression of RANKL,while the former is better;titanium could significantly increase the expression of RANKL,while PLA couldn't increase compared with control group.Conclusion PLA has less influence on cell cytotoxicity and the expression of OPG/RANKL mRNA than Titanium,NiCr and CoCr.
