卒中后抑郁大鼠小脑功能改变的实验研究

摘　　要：目的观察卒中后抑郁(post-stroke depression,PSD)模型大鼠小脑功能的改变。方法选用健康雄性SD大鼠,随机分为4组,对照组、卒中组、单纯抑郁组、PSD组;分7d、14d、21d 3个时间进行行为学观察和测试、免疫组化法测定小脑组织功能活性依赖蛋白突触素(P38)、脑源性神经营养因子(BDNF)的表达。结果与卒中组相比较,单纯抑郁组大鼠的体重及糖水消耗量均下降,水平运动和垂直运动水平均降低(P<0.01);PSD组大鼠体重及糖水消耗量下降程度更为明显,水平运动和垂直运动水平降低程度最为明显(P<0.01)。与卒中组比较,单纯抑郁组P38、BDNF的表达普遍减弱;与单纯抑郁组相比较,PSD组P38、BDNF的表达减弱的更为明显;组内(7d、14d、21d)比较:卒中组、单纯抑郁组、PSD组随时间推移,小脑组织P38、BDNF表达呈现减弱趋势。结论大鼠小脑功能改变可能与PSD发生有关,进一步研究可能有助于阐明卒中后抑郁症的发病机制。【Objective】 To observe the changes of cerebellar function in the post-stroke depression（PSD） model of rats.【Methods】The healthy SD rats were randomLy divided into four groups,control group,stroke group,depression group,and post-stroke depression（PSD）group.Behavioral observation and testing were performed and the expression P38 and BDNF in cerebellum by immunohistochemical method of 7days,14days and 21days are detected 【Resluts】 Compared with stroke group,depression rats,weight and sugar water consumption are decreased,horizontal motion and vertical motion level are lower（P 0.01）;PSD rats,weight and sugar water consumption drop are more apparent,horizontal motion and vertical motion level also reduce the most obvious（P 0.01）.Group comparison：Compared with the stroke group,depression group and PSD group,the expression of P38 and BDNF in cerebellum are decreased.The difference between them was very significant.Intergroup comparison（7d,14d,21d）：Compared with the stroke group,depression group and PSD group,P38 and BDNF were significant difference.【Conclusion】 Cerebellar dysfunction has the potential to be involved in PSD.
