补肾益肺消癥方对肺纤维化大鼠MMPs基因表达及病理改变的影响  被引量:3

摘  要:目的探讨补肾益肺消癥方治疗特发性肺纤维化的作用机制。方法将SD大鼠随机分为空白组、阴性对照组、阳性对照组、中药小剂量组、中药中剂量组、中药大剂量组,以博莱霉素致大鼠肺纤维化模型。阳性对照组给予地塞米松灌胃,空白组、阴性对照组分别给予等量生理盐水,中药小、中、大剂量组分别按成人剂量组5,10,20倍灌胃。通过Real-time PCR检测各组肺组织中基质金属蛋白酶(MMP)-2、MMP-9基因表达情况,HE染色观察大鼠肺组织病理的改变。结果模型组大鼠肺组织中MMP-2基因表达量相比其他各组明显升高,中药治疗组表达量与空白组相近;中药治疗组大鼠肺组织中MMP-9明显高于模型组(P<0.01);肺组织切片HE染色分析结果显示,阳性对照组和中药治疗组肺泡壁轻度增厚,肺泡结构破坏程度较轻,存在成纤维细胞增生,纤维化程度较模型组明显改善。结论补肾益肺消癥方可以缓解免疫应答所导致的炎症反应及细胞外基质蛋白和胶原的沉积,从而抑制肺组织纤维化的进程,达到治疗效果。Objective It is to explore the mechanism of Bushenyifeixiaozheng decoction in the treatment of idiopathic pulmonary fibrosis(IPF).Methods SD rats were randomly divided into blank group,negative control group,positive control group,low,moderate and high dosage of Chinese drug groups,IPF rat models were caused by Bleomycin.The positive control group was given Dexamethasone by intragastric administration,blank group and negative group were given the same dosage of normal saline,the Chinese drug groups were given corresponding dosages of Bushenyifeixiaozheng decoction.The gene expressions of matrix metalloproteinase(MMP) 2,9 in lung of IPF rats were detected by real-time PCR.The pathological changes in lung of the rats were observed through hematoxylin dyeing.Results The gene expression of MMP-2 in lung tissue in model group was obviously higher than that in the other groups,and the expression in Chinese drug treatment groups was similar to the blank group.The gene expression of MMP-9 in lung tissue in Chinese drug treatment group was obviously higher than that in model group.The result of HE stain analysis in lung tissue showed that Alveolar wall in rat's lung of positive control group and Chinese medicine treatment groups thickened mildly,fibroblasts proliferation existed,the level of fibrosis was improved obviously compared with that of model group.Conclusion Bushenyifeixiaozheng decoction can relieve the inflammatory reaction and inhibit deposition of extracellular matrix protein and collagen in diseased region to refrain the process of pulmonary fibrosis.This should be the key mechanism of IPF treated by this decoction.
