H_2S促进神经胶质瘤细胞增殖作用的机制探讨

摘　　要：目的为了明确硫化氢(H2S)是否能够促进人胶质瘤U251细胞的增长,探讨Survivin和Sirt1在U251细胞中是否有表达及H2S促进U251细胞增长机制。方法 CCK8(Cell Counting Kit-8)法测细胞活力,Western Blot检测U251细胞内Survivin和Sirt1表达。结果 100-400μmol/L的H2S呈浓度依赖性地促进了U251细胞的增长;100-400μmol/L的H2S处理U251细胞24 h后,细胞内Survivin和Sirt1呈H2S浓度依赖性地增加,与空白对照比较,差异有统计学意义(P<0.01);用100μmol/L Sirt1抑制剂,Sirtinol预处理U251细胞0.5 h后,在继续用200μmol/L H2S继续培养细胞4 h后,预处理组和单独的硫化氢组相比,细胞的活力显著下降,差异有统计学意义(P<0.001),但是单独Sirtinol组对细胞活力影响不大,说明H2S促进细胞增殖功能可能是通过促进Sirt1的表达来实现的。结论 H2S促进了U251细胞的增长,H2S促进了U251细胞内Survivin和Sirt1表达,Sirt1抑制剂抑制了H2S对细胞的促进增长作用。Objective To explicite whether hydrogen sulfide（H2S） could promote the growth of human glioma U251 cells,to investigate the expression of Sirt1 and Survivin in U251 cells and to explore the mechanism of H2S for promoting the U251 cells growth.Methods The cell counting kit-8（CCK8） assay was used to measure the cell vitality.The expressions of Sirt1 and Survivin were detected by Western blot.Results 100-400 μmol/L H2S promoted the growth of U251 cells by the concentration dependency mode.After treating U251 cells by 100-400 μmol/L H2S,the expressions of intracellular Sirt1 and Survivin were concentration-dependently increased（P0.01）.After treating U251 cells by Sirt1 inhibitor Sirtinol（100 μmol/L） for 30 min and successively culturing U251 cells by 200 μmol/L H2S for 4 h,comparing the pretreatment group with the single H2S group demonstrated that the cell vitality was significantly decreased with statistical difference（P0.001）.But the single Sirtinol group seldom affected the cell vitality,which indicating that the role of H2S promoting the cell proliferation could be realized by promoting the Sirt1 expression.Conclusion H2S increases the growth of U251 cells,at the same time,improves the intracellular level of Sirt1 and Survivin.The inhibitor of Sirt1 inhibits the promotion role of H2S on U251 cells.
