摘 要:目的:探讨蛇毒精氨酸酯酶Agkihpin对人肝癌SMMC-7721细胞活力、增殖和迁移的影响。方法:用不同浓度的Agkih-pin处理肝癌细胞株后,应用MTT法检测细胞活力,应用细胞计数法检测Agkihpin对细胞增殖和迁移的影响。结果:0.207~4.130mg/L浓度的Agkihpin作用SMMC-7721细胞后,与对照组比较,各加药组细胞活力明显下降(P<0.01),细胞活力随Agkihpin浓度增加而减弱(r=-0.81,P<0.05),细胞活力抑制率可达12.2%~46.2%;2.065~4.130mg/L剂量的Agki-hpin对人肝癌SMMC-7721细胞的增殖有明显的抑制作用(P<0.05),加药24,48h和72h后增殖抑制率分别为29.4%~88.2%、64.1%~84.6%和4.4%~30.5%,且剂量依赖作用明显(P<0.05);3.089mg/L和4.130mg/L剂量的Agkihpin对SMMC-7721细胞还有一定的杀伤作用,杀伤率分别为20.0%和79.6%;0.207~4.13 mg/L剂量的Agkihpin对人肝癌SMMC-7721细胞的迁移有明显的抑制作用(P<0.01),细胞迁移抑制率可达7.8%~98.0%,且剂量越大抑制作用越明显(r=0.841,P<0.05)。结论:一定剂量的Agkihpin可抑制人肝癌SMMC-7721细胞株的细胞活力、增殖和迁移。Objective: To explore the impacts of Agkihpin on the cellular vitality, proliferation and migration of hepatocellular carcinoma SMMC-7721 cell strain. Methods: The cultured SMMC-7721 cells were treated with different concentrations of Agkihpin, then the cellular vitality was detected by MTT, the impacts on proliferation and migration of cell were detected by cell count. Results: Compared with the control group, the cellular vitality of SMMC-7721 was reduced obviously ( P 〈0.01) with the increasing of concentration of Agkihpin ( P 〈0.05, r =-0.81), and the inhibiting rate ranged from 12.2%-46.2% after the treatment of 0. 207-4. 130 mg/L Agkihpin; the proliferation of SMMC-7721 was inhibited obviously in a concentration-depended manner ( P 〈0.05), and the inhibiting rate ranged from 29.4%-88.2%, 64. 1%-84.6% and 4.4%-30.5% respectively, after treated with 2. 065-4. 130 mg/L Agkihpin for 24 h, 48 h and 72 h; the concentration of 3. 089 and 4. 130 mg/L Agkihpin can cause casualty of SMMC-7721 in a rate of 20.0% and 79.6%; the migration of SMMC-7721 was reduced obviously ( P 〈0.01) with the increasing of concentration of Agkihpin (P 〈0.05, r =-0. 841), and the inhibiting rate ranged from 7. 28%-98.0%. Conclusion: Agkihpin can inhibit the cellular vitality, proliferation and migration of SMMC- 7721 cells significantly. Agkihpin may work effectively in therapy and prevention of hepatocellular carcinoma in future.
