摘 要:目的研究鸡血藤总黄酮对大鼠脑缺血的保护作用及其作用机制。方法采用线栓法制备大鼠大脑中动脉缺血模型,观察鸡血藤总黄酮对局灶性脑缺血大鼠行为障碍、脑梗死面积的影响;采用结扎双侧颈总动脉法制备大鼠急性脑缺血再灌注模型,观察鸡血藤总黄酮对急性脑缺血大鼠脑含水量以及脑组织SOD、GSH-Px酶活性和MDA含量的影响。结果鸡血藤总黄酮(40 mg/kg、80 mg/kg)灌胃给药明显改善大脑中动脉栓塞致脑缺血大鼠的行为障碍,减少缺血区面积,同时能减轻急性脑缺血再灌注损伤引起的大鼠脑水肿,抑制缺血致脑组织SOD、GSH-Px活性的降低和MDA水平的升高。结论鸡血藤总黄酮对大鼠实验性脑缺血具有一定的保护作用,其机制可能与通过提高机体抗氧化能力有关。Objective To investigate the protective effect and mechanism of suberect spatholobus total flavonoids(SSTF) on mice with cerebral ischemia.Methods The mice mode with ischemia on middle cerebral artery was made by suture-occluded method.The effect of SSTF on behavior disorder state and cerebral infarction area of mice with focal cerebral ischemia was observed.The mice mode with acute cerebral ischemia reperfusion was made by the method of ligaturing bilateral carotid artery occlusion.The effect of SSTF on cerebral water content and SOD、GSH-Px' enzyme activity and MDA content of mice with acute cerebral ischemia reperfusion was observed.Results Intragastric administration with SSTF(40、80 mg/kg)improved obviously behavior disorder state of cerebral ischemia mice mode with middle cerebral artery embolism and reduced ischemic region area and relieved encephaledema level of mice with acute cerebral ischemia reperfusion,suppressing reduction of SOD、GSH-Px' enzyme activity and rise of MDA content in ischemic brain tissue.Conclusions SSTF has certain protective effect on mice mode with experimental cerebral ischemia,which may be relevant to SSTF capacity of increasing body resistance to oxidation.
