摘  要:目的:探讨中药银杏叶提取物(EGb761)对血管性痴呆(vascular dementia,VD)模型大鼠海马齿状回(DG)NSCs增殖分化的影响及机制。方法:采用双侧颈总动脉夹闭再灌注同时腹腔注射硝普纳建立VD模型,在15 d,1,2,4个月等时间点,采用Morris水迷宫检测和BrdU和/或神经颗粒素(Ng)标记的免疫荧光单标和双标技术分别观察大鼠的学习记忆能力变化和NSCs的增殖与功能分化情况。结果:(1)模型组大鼠的逃逸潜伏期(EL)均明显长于假手术组(P<0.01),药物组大鼠的EL较模型组明显缩短(P<0.05),但仍长于假手术组(P<0.01)。(2)免疫荧光标记显示,各个时间点,药物组DG区NSCs在增殖分化的数量、强度和持续时间上,都明显优于假手术组和模型组。结论:EGb761能促进VD模型大鼠神经干细胞的增殖分化,显著改善VD大鼠的学习记忆能力。Objective: To explore the effects of EGb761 on the proliferation and differentiation of neural stem cells in dentate gyrus (DG) of vascular dementia(VD) rats. Methods: VD models were established by repeatedly clipping the common carotid arteries of the rat in combination with an intraperitoneal injection of sodium nitroprusside solution in anes- thetized SD rats. Morris Water Maze test was used to detect the learning and memory ability of the rats and immunofluo- rescence single and double scale method to detect the proliferation and differentiation of neural stem cells on 15 d, 1,2, 4 months time points. Results: ( 1 ) The escape latency (EL) in the VD group was significantly longer than that in the sham-operated group (P 〈 0.01 ), but that in the treatment group was shorter than that in the VD modal group (P 〈 0.05 ) , and longer than in the sham group ( P 〈 0.01 ). (2) lmmunofluorescence staining showed that the amount, inten- sity and duration in the DG of proliferation and differentiation of treatment group was significantly higher than that in the sham-operated group and the VD group. Conclusion: Treatment of EGb761 could enhance the proliferation and differenti- ation of neural stem cells in VD rats, and improve obviously the learning and memory abitity of VD rats.
