摘  要:对犬瘟热病毒(CDV)PS株基因组进行全基因组序列测定与分析,以阐明该毒株的基因特征与遗传进化情况。利用RT-PCR方法分段扩增PS株全基因组序列,分别将产物克隆入pMD18-T载体中并进行测序,将所测序列拼接获得CDV PS株的全基因组cDNA。用DNAStar软件比较PS株全基因组序列与国内外犬瘟热病毒疫苗株和野毒株全基因组序列的同源性,然后用Mega4.0软件进行系统进化分析。测序结果显示,PS株全基因组序列的长度为15 690nt,该基因组推导的某些氨基酸位点发生了突变。与GenBank中登录的国内外参考毒株的同源性比较结果显示,PS株基因组与MKY-KM08分离株的基因序列同源性高达97.9%,与其他毒株的同源性为93.0%~97.0%;与CDV3疫苗株和Onderstepoort疫苗株的同源性最低,分别为93.2%和93.0%。基于全基因组序列绘制的系统进化树表明,所有CDV分离株分为疫苗株和野毒株2支,PS株和MKY-KM08株位于同一支,属于亚洲1型。结果表明PS株为野毒株,其全基因组序列与MKY-KM08株的全基因组序列亲缘关系最近。To sequence the full genome of canine distemper virus(CDV) PS strain and to study the genetic variation and molecular features of the PS strain, 10 fragments of the full length genome of CDV PS strain were amplified by RT-PCR,cloned into the pMD18-T vector and sequenced. The homology and evo-lutional relationship of CDV genome with reference strains were analyzed with DNAStar and Mega4.0 soft-ware. Sequence analysis showed that the complete genome was 15 690 bp in length with some amino acid site mutated. The homology analysis showed that the PS strain had a 97.9 % similarity with MKY-KM08 strain,and 93.0G to 97.0; with other strains. The PS strain had a similarity of 93.2; and 93.0; with CDV3 and Onderstepoort vaccine strains, respectively. Phylogenetic analysis showed that all CDV strains included vaccine strain and wild strain. PS strain and MKY-KM08 strain both belonged to Asia-1 wild strain. The results indicated that the genome sequence of the PS strain had highest similarity with that of the MKY-KM08 strain,and it was a wild strain.
