多烯磷脂酰胆碱对大鼠非酒精性脂肪性肝病的防治作用观察

摘　　要：目的观察多烯磷脂酰胆碱对大鼠非酒精性脂肪性肝病(NAFLD)的预防作用。方法选用Wistar大鼠48只,随机分为正常组(14只)、模型组(14只)、预防组(10只)和治疗组(10只)。模型组大鼠在高脂饮食喂养12周成功诱导出NAFLD模型。预防组提前应用多烯磷脂酰胆碱胶囊干预4周,再加用高脂饮食处理12周;治疗组给予模型动物多烯磷脂酰胆碱胶囊治疗12周。采用Western Bloting法检测肝组织葡萄糖调节蛋白78(GRP78)和固醇调节元件结合蛋白1-c(SREBP-1c)表达。结果在12周时,与模型组比较,预防组和治疗组大鼠肝脏脂肪变程度减轻,血清ALT、AST、TG、FFA水平显著下降(P<0.01);正常组肝组织GRP78和SREBP-1c表达量分别为12.35±0.50和10.09±0.52,而模型组分别为40.67±1.72和34.67±1.54,预防组分别为17.97±0.89和15.73±0.58,治疗组分别为18.07±0.81和17.82±3.28(P<0.01);预防组与治疗组间以上各个指标比较,差异无统计学意义。结论多烯磷脂酰胆碱胶囊对单纯高脂饮食诱导的NAFLD大鼠有治疗作用,但预防性用药无明显效果。Objective To observe the effect of polyene phosphatidylcholine on rats with nonalcoholic fatty liver disease （NAFLD） . Methods 48 Wistar rats were randomly divided into normal （n=14）,model（n=14）,prophylactic （n=10） and treatment group（n=10）. Rats in prophylactic group were fed with polyene phosphatidylcholine for 4 weeks before model preparation. The expression of GRP78 and SREBP-lc in liver tissues were detected by Western bloting. Results The hepatic steatosis in prophylactic and treatment group was reduced obviously as compared to in model group;the serum levels of AST,ALT,TG and FFA were significantly reduced in prophylactic and treatment group （P〈 0.01）;the expression of GRP78 and SREBP-1c in liver tissues were 12.35±0.50 and 10.09±0.52 in normal group, 40.67±1.72 and 34.67±1.54 in model group,17.97±0.89 and 15.73±0.58 in prophylactic group and 18.07±0.81 and 17.82±3.28 in treatment group （P〈0.01）;However,all indexes mentioned above between prophylactic and treatment group had no statistical difference. Conclusions Polyene phosphatidylcholine has a certain therapeutic effect on rats with NAFLD induced by high fat diet feeding, and prophylactic application of polyene phosphatidylcholine do not enhance the curative effect.
