磷酸化PRAS40蛋白在膀胱尿路上皮癌中的表达及其临床意义

摘　　要：目的探讨膀胱尿路上皮癌组织中磷酸化PRAS40蛋白的表达及其临床意义。方法应用量子点免疫荧光组织化学方法检测76例膀胱尿路上皮癌（BUC）和20例癌旁正常膀胱组织中磷酸化PRAS40蛋白的表达，并分析其与临床病理特征的关系。结果磷酸化PRAS40蛋白在膀胱尿路上皮癌组织中表达阳性率为51．3％（39／76），在20癌旁正常膀胱组织中表达均为阴性。在低度恶性倾向尿路上皮乳头状瘤（PUNLMP）及低分级尿路上皮癌组织中表达阳性率为36．8％（14／38），而在高分级癌组织中的阳性率为65．8％（25／38），两组比较，差异有统计学意义（P〈0．05）；在非肌层浸润性膀胱尿路上皮癌和肌层浸润性膀胱尿路上皮癌组织中的阳性率分别为40．0％（16／40）和63．9％（23／36），差异有统计学意义（P〈0．05），而其表达水平与肿瘤患者年龄、肿瘤数目大小、初复发情况等无明显相关（P〉0．05）。结论磷酸化PRAS40蛋白的表达与膀胱尿路上皮癌的恶性进展密切相关。Objective To investigate the exPression of phosphorylated PRAS40 in bladder urothe- lial cancer and its clinical significance. Methods The expression of phosphorylated PRAS40 in 76 cases of bladder urothelial cancer and 20 cases of normal bladder tissue was examined by using quantum-dot-based immunofluorescence histochemistry, and its correlations to clinicopathological parameters were analyzed. Results The positive expression rate of phosphorylated PRAS40 in bladder urothelial cancer was higher than in the normal bladder tissue （51.3% versus 0% ）. The positive expression rate of phosphorylated PRAS40 was 36. 8% and 65.8% in PUNLMP plus low-grade BUC and high-grade BUC （P 〈0. 05）, while 40. 0% and 63.9% in NMIBUC and MIBUC （P 〈0. 05）, respectively, but no significant association was identified between phosphorylated PRAS40 expression and gender, tumor size, tumor number, recurrence. Conclusion Phosphorylated PRAS40 is closely correlated with the malignant progression of bladder urothelial cancer. Phosphorylated PRAS40 may be a robust biomarker for evaluating the prognosis and the effectiveness of molecule targeted therapy in BUC.
