摘  要:目的观察肾透明细胞癌(ccRCC)与正常肾组织中ALS2CL基因的表达及差异,并探讨其意义。方法采用实时定量聚合酶链反应(PCR)、免疫组织化学(IPH)及Westernblot检测26例配对癌组织/癌旁正常肾组织标本及2株ccRCC来源细胞(786/0、SN12-PM6)中ALS2CL基因mRNA和蛋白的表达。结果比较正常对照组,ALs2CL基因mRNA在65.38%(17/26)的eeRCC组织中表达显著下调,在786/0、SNl2一PM6中表达亦显著下调;IPH结果示73.08%(19/26)ccRCC组织ALS2CL蛋白呈阴性或弱阳性表达;WesternNot结果显示53.58%(14/26)ccRCC组织ALS2CL蛋白表达下调,两株细胞ALS2CL蛋白表达亦下调,比较各自对照组,其表达差异均有统计学意义(P〈0.05)。结论ALS2CL基因表达缺失或下调可能与肾透明细胞癌发生密切相关。Objective To study the differential expression of ALS2CL in clear cell renal cell car- einoma (eeRCC) and normal kidney tissue, and its implication. Methods Real-time polymerase chain re- action (PCR), immunohistoehemistry and Western blotting were applied to examine the expression of ALS2CL in 26 cases of ceRCC tissues and adjacent normal kidney tissues as well as two kinds of ceRCC- derived cell lines (?86/0, SN12-PM6). Results As compared with normal control, the two ccRCC-de- rived cell lines (786/0, SN12-PM6) and 65.38% (17/26) of ceRCC tissues showed an obvious down- regulated expression of ALS2CL mRNA. IPH revealed that 73.08% ( 19/26 ) of ccRCC specimens were negative or weakly positive for ALS2CL protein. Western blotting also showed a highly down-regulated rate of 53.85% (14/26) in ccRCC samples, and the low expression of ALS2CL was also found in 786/0 and SN12-PM6 cells. All of them showed significant difference between ceRCC and corresponding normal con- trol, P 〈 0. 05. Conclusion The absence or downregulated expression of ALS2CL may be closely related to the occurrence of ccRCC.
