摘 要:目的观察雷公藤红素对人类原代乳腺癌细胞及乳腺癌干细胞的毒性效应,探讨其在人类乳腺癌治疗中的作用。方法体外培养人类原代乳腺癌细胞及干细胞,施加不同浓度的雷公藤红素,0.1、0.5、1.0μmol/L,观察其被杀伤及凋亡的结果。结果雷公藤红素对人乳腺癌细胞及乳腺癌干细胞都具有诱导凋亡的作用,而且对乳腺癌细胞的毒性效果更加明显。雷公藤红素为1μmot/L时,72h后乳腺癌细胞实验组凋亡比例接近60%,乳腺癌干细胞实验组凋亡比例也超过30%,其凋亡比例都显著高于对照组(P〈0.05)。结论雷公藤红素能有效诱导人乳腺癌细胞及其干细胞凋亡。Objective To investigate the efficacy of tripterine for the treatment of the human breast cancer through the observation of cytotoxicity on human primary breast cancer cells and the stem cells in vitro. Methods Human primary breast cancer cells and breast cancer stem ceils from breast invasive ductal carcinoma samples were cultured and inoculated with tripterine of three different concentrations (0. 1, 0. 5, 1.0 μmol/L). The cell killing and apoptosis were observed every day. Results Tripterine was cytotoxic for human primary breast cancer cells and breast cancer stem cells in vitro, but the cytotoxicity for human primary breast cancer cells was stronger than the stem cells. In the presence of tripterine 1 p^mol/L after co-culture for 72 h, the apoptotie rate of human primary breast cancer cells and the stem cells was approximately 60% and over 30% , respectively. The apoptosis rate in experimental group was significantly higher than in control group ( P 〈 0. 05 ). Conclusion Tripterine can effectively induce apoptosis of human primary breast cancer cells and breast cancer stem cells in vitro.
