摘  要:目的观察黄芪对HSC—T6细胞基质分解素-1(MMP3)及其抑制因子基因表达的影响。方法以不同浓度的黄芪(1.0、0.5g/L)作用于培养的肝星状细胞株HSC—T6细胞48h,收集细胞,提取总RNA;用逆转录定量聚合酶链反应(RT—PCR)方法测定MMP3及金属蛋白酶组织抑制因子-1(TIMP-1)的基因表达水平。结果MMP3基因表达水平在黄芪1.0、0.5g/L两组分别为3.36±0.42、2.63±0.36,而空白对照组为2.13±0.30。TIMP.1基因表达水平在黄芪1.0、0.5g/L两组分别为3.96±0.66、4.13±0.50,而空白对照组为4.03±0.56。结论黄芪可明显增强HSC—T6细胞MMP3的基因表达,且与剂量有关;但对TIMP-1的基因表达无明显影响。Objective To investigate the effects of Astragalus on gene expression of stromelysin-1 (MMP3) and tissue inhibitor of metalloproteinase-1 (TIMP-1) in HSC-T6 cells. Methods HSC-T6 cells were cultured in medium containing different concentrations of Astragalus ( 1.0, 0. 5 g/L) for 48 h and harvested. Total RNA of HSC-T6 cells was isolated and gene expression levels of MMP3 and tIMP-1 were measured by using reverse transcription polymerase chain reaction. Results MMP3 gene expression level in different groups of Astragalus ( 1.0, 0. 5 g/L) was 3.36 ± 0. 42, and 2. 63 ± 0. 36 respectively, and that in control group was 2. 13 ± 0. 30. TIMP-1 gene expression level in different groups of Astragalus ( 1.0, 0. 5 g/L) was 3.96 ± 0. 66, and 4. 13 ± 0. 50 respectively, and that in control group was 4. 03±0. 56. Conclusion Astragalus may significantly increase MMP3 gene expression of HSC-T6 cells in a dose- dependent manner. Astragalus has no significant effect on TIMP-1 gene expression.
