摘  要:目的观察柴胡对HSC—T6细胞基质分解素-1(MMP3)及I型胶原基因表达的影响。方法以不同浓度的柴胡(1.0、0.5g/L)作用于培养的肝星状细胞株HSC—T6细胞48h,收集细胞,提取总RNA;用逆转录定量聚合酶链反应(RT—PCR)方法测定MMP3及I型胶原的基因表达水平。结果MMP3基因表达水平在柴胡1.0、0.5g/L两组分别为2.02±0.29、2.30±0.33,而空白对照组为2.13±0.30。I型胶原基因表达水平在柴胡1.0、0.5g/L两组分别为0.82±0.13、1.11±0.13,而空白对照组为3.12±0.46。结论柴胡可明显抑制HSC—T6细胞I型胶原的基因表达,且与剂量有关;但对基质分解素-1的基因表达无明显影响。Objective To investigate the effects of Radix Bupleuri on gene expression of stromely- sin-1 (MMP3) and type I collagen in HSC-T6 cells. Methods HSC-T6 cells were cultured in medium containing different concentrations of Radix Bupleuri (1.0, 0. 5 g/L) for 48 h and harvested. Total RNA of HSC-T6 cells was isolated and gene expression levels of MMP3 and type I collagen were measured by using reverse transcription polymerase chain reaction. Results MMP3 gene expression level in different groups of Radix Bupleuri ( 1.0, 0. 5 g/L) was 2. 02 ±0. 29 and 2. 30 ±0. 33 respectively, and that in con- trol group was 2. 13 ±0. 30. Type I collagen gene expression level in different groups of Radix Bupleuri (1. 0, 0. 5 g/L) was 0. 82 ± 0. 13 and 1.11 ± 0. 13 respectively, and that in control group was 3.12 ± 0. 46. Conclusion Radix Bupleuri may significantly inhibit type I collagen gene expression in HSC-T6 cells in a dose-dependent manner. Radix Bupleuri has no effect on MMP3 gene expression.
