摘  要:目的通过对人食管鳞癌EC9706细胞线粒体DNA(mtDNA)中ND1基因进行检测,分析其基因突变的意义。方法培养人食管鳞癌EC9706细胞,提取其mtDNA中的ND1基因,并测序。结果测序发现,人食管鳞癌EC9706细胞mtDNA中ND1基因的3 971处出现点突变,由C突变为T。结论人食管鳞癌EC9706细胞中mtDNA的ND1基因的突变与食管癌发生、发展关系密切。Objective To study the mutation of the mitochondrial DNA (mtDNA) ND1 gene of the human e- sophageal squamous carcinoma EC9706 cells. Methods The mtDNA was extracted in human esophageal squamous carcinoma EC9706 cells and ND1 gene was amplified and sequenced. Results The gene sequencing was success- fully detected. Point mutation ( C3 971T) in ND1 gene was found by sequencing in the human esophageal squamous carcinoma EC9706 cells. Conclusion The mutation of the mtDNA ND1 gene of the human esophageal squamous carcinoma EC9706 ceils is related to the occurrence and development of esophageal carcinoma.
