摘  要:目的:建立多药耐药人胃癌细胞裸鼠移植瘤模型并探讨移植瘤的耐药特性。方法:将人胃癌细胞株SGC-7901和耐药细胞株SGC-7901/HCPT分别接种于裸小鼠左侧背部皮下和右侧背部皮下,在SPF级动物合格环境下饲养,观察肿瘤细胞成瘤情况,用MTT法比较移植瘤细胞对羟基喜树碱的敏感性及免疫组化测定其耐药基因TOPO-Ⅰ表达的变化。结果:SGC-7901和SGC-7901/HCPT裸小鼠移植成瘤率为100%,无自行消退现象。MTT法测定SGC-7901/HCPT和SGC-7901/HCPT/nude的IC50分别为4.686μg/mL和4.381μg/mL,与移植前细胞比较,差异无统计学意义。耐药基因TOPO-Ⅰ表达分别为2.964±0.162和2.953±0.172,差异无著性意义。结论:以SGC-7901/HCPT细胞建立的裸鼠皮下耐药移植瘤模型,仍保持体外的耐药活性和基因表型,为耐药机制和逆转的研究提供了良好的动物模型。Objective:To establish multi-drug resistance human gastric cancer cell hetero-transplanted models in nude mice and to ex- plore its multi-drug resistance characteristics. Methods: Human gastric cancer cell line SGC-7901 and resistant cell lines SGC-7901/ HCPT were inoculated in the left and right subcutaneous part of these nude mouse respectively reared in the SPF level of qualified animal environment. Observed transplanted tumor' growth was compared with the MTT method HCPT tumor cell sensitivity and immunohisto- chemical determination of the resistance gene expression in TOPO-I. Results: SGC-7901 and SGC-7901/HCPT nude mice tumor forma- tion rate was 100% with no self-limiting phenomenon. The growth rate showed no significant difference. Determination SGC-7901/HCPT and SGC-7901/HCPT/nude MTT,ICS0 4. 686 μg/mL and 4. 381 μg/mL,compared with the ceils before transplantation showed no signif- icant difference,resistance gene expression by TOPO-I was 2. 964 ± 0. 162 and 2. 953 ± 0. 172. The difference was not significant meaning. Conclusion: The SGC-7901/HCPT cells established in nude mice xenograft model of subcutaneous resistance had resistance remaining active and in vitro phenotype for the reversal of drug resistance mechanisms and provide a good animal model.
